摘  要:目的 :观察非程控降温 - 80℃冷冻保存外周血造血干 /祖细胞的活性 ,为临床自体外周血造血干细胞移植提供方便、有效的冻存方法。方法 :常规方法动员外周血干细胞 ,CS - 30 0 0PLUS血细胞分离机采集干细胞 ,以 10 %二甲亚砜、10 %人AB型血清、RPM116 4 0培养基为冷冻保护剂 ,2ml/管冻存干细胞 ,按第 1、2、4周和第 2、3月顺序复苏细胞并测定GM -CFU集落数、台盼兰拒染率和CD34+ 细胞百分率。结果 :用该方法冷冻保存的外周血干细胞 ,在 4周内其细胞活性与采集时的细胞活性无明显差别 (P >0 .0 5 ) ,冻存至 2~ 3个月时细胞活性则明显降低 (P <0 .0 5 )。结论 :应用血清和DMSO等作为冷冻保护剂、- 80℃直接冻存的外周血干细胞 ,在 4周内复苏后其细胞活性无明显降低。Objective:To observe the activity of peripheral blood stem cells cryopreserved at-80℃ without rate-controlled freezing and provide an convenient, effective freezing and preserving method for PBSCT. Methods:We mobilized and collected peripheral blood stem cells in common methods,froze cells with 10% DMSO,10% human serum of AB type and RPMI Medium 1?640 without rate-controlled,and preserved them at-80℃.We recovered cells in 1th,2nd,4th week and 2nd,3th months,and detected the cell activity with some methods such as the number of GM-CFU, percent of CD34 + cells and trypan blue exclusion. Results:There was no difference between frozen cells and collected cells in activity within 4 weeks( P >0.05), whereas there was some obvious difference between frozen cells and collected cells after frozen for 2 and 3 months ( P >0.05). Conclusions:The peripheral blood stem cells may be frozen at-80℃ without rate-controlled freezing and their activity may not be obviously changed within 4 weeks.
