摘  要:为了建立一个大豆子叶节高效再生体系用于大豆的遗传转化 ,选用 10个东北主栽大豆品种(Glycinemax (L .)Merr .)的子叶节作为外植体 ,研究了基因型、植物激素的浓度、超声波辅助处理时间 ...等 6种影响大豆子叶节再生的因素。结果表明合丰 35、合丰 2 5、黑农 4 0再生效率较高 ,平均每个外植体分化出的丛生芽数分别为 6 .6 9、6 .32和 5 .98;确定了丛生芽分化阶段所需的BA浓度和生根阶段所需的IBA浓度 ,发现不同基因型需要不同的BA浓度 ,合丰 35、合丰 2 5为1.5mg/l,黑农 4 0为 1.2mg/l。适宜的外植体大小为保留全部子叶。作为遗传转化时的除菌剂 ,头孢唑啉钠 (Cef)在 5 0 0mg/l时对子叶节的再生无显著影响。当遗传转化时使用的超声波辅助处理时间小于 12秒时 。To establish a high frequency regeneration system for the gene transformation of soybean(Glycine max (L.) Merr.), cotyledonary nodes from ten varieties were used as the explants in the regeneration experiments. Six factors such as genotypes, plant growth regulators ,sonication assisted treatment and so on were studied. The results show that the high regeneration frequency genotypes are Hefeng 35, Hefeng 25,Heinong 40, the multiple shoots per explant are 6.69,6.32 and 5.98 respectively. The BA concentrations required in shoot differentiation medium are different among these genotypes, in Hefeng 35, 1.5mg/l Hefeng 25 and in 1.2mg/l Heinong 40. But the IBA concentrations in root regeneration medium are the same. The full cotyledonary is the best explant size for shoot differentiation. As an antibiotic to restrain the propagation of excess Agrobacterium ,the Cef has no significant influence on regeneration. When the treatment time is shorter than 12s , the sonication assisted treatment has no significant influence on the regeneration of shoots.
