摘  要:目的:探索日本血吸虫单克隆抗独特型抗体NP30对血吸虫虫卵肉芽肿细胞凋亡的影响。方法:BALB/c小鼠随机分为两组,实验组腹腔注射NP30主动免疫3次,对照组腹腔注射生理盐水,分别在尾蚴攻击感染后第39、49、64、108、112天处死,应用透射电镜观察肝虫卵肉芽肿细胞凋亡的形态改变,流式细胞术(FCM)和末端转移酶介导的缺口末端标记法(TUNEL)检测虫卵肉芽肿凋亡细胞。结果:透射电镜观察虫卵肉芽肿细胞有凋亡小体形成;经FCM和TUNEL检测实验组虫卵肉芽肿细胞凋亡明显多于对照组。结论:细胞凋亡在日本血吸虫虫卵肉芽肿的形成、细胞转化过程中起着重要作用。NP30抗病免疫作用的机制之一是促进虫卵肉芽肿细胞的凋亡。Objective:To study the effect of monoclonal anti-idiotypic antibody NP30 on apoptosis of egg granuloma cells in mice infected with Schistosoma japonicum. Methods:BALB/c mice were divided at random into two groups. The mice of the experimental group were immunized by injecting NP30 intraperitoneally for three times,while control group was injected with normal saline intraperitoneally. The mice were killed at the 39th、49th、64th、108th、112th day after being challenged with cercariae.The morphology of apoptotic cells of egg granuloma in mice liver were studied by electron-microscopy,and the quantity of apoptotic cells was evaluated by TdT mediated dUTP nick end labeling(TUNEL)and Flow Cytometry(FCM).Results:Apoptotic body was seen to have been formed in egg granuloma cells under electron-microscopy,apoptotic egg granuloma cells of the expeimental group were much more than those of the control group.Conclusion:The apoptosis of egg granuloma cells plays an important role in the formation of granuloma of Schistosoma japonicum.One of anti-pathology immunity mechanism of NP30 is to promote of egg granuloma cells.
