急性胰腺炎局部微循环中性粒细胞内[Ca^(2+)]_i变化与细胞粘附分子表达的调控及其意义

摘  要:目的 探讨急性胰腺炎 (AP)早期大鼠胰腺局部微循环中性粒细胞在流体切应力作用下细胞内 [Ca2 + ] i变化与表面粘附分子CD18和CD62L表达的调控及其意义。方法 将Wistar大鼠随机分为 3组 (n =7) :正常组、实验Ⅰ组 (AP 2h组 )和实验Ⅱ组 (AP 4h组 ) ;用蛙皮缩胆囊肽 (caerulein)诱发大鼠AP动物模型。用流式细胞仪分析切应力作用下胰腺局部微循环中性粒细胞 [Ca2 + ] i的变化、CD18和CD62L的表达。结果 低切变率作用下各组 [Ca2 + ] i 呈现轻微下调 ,以后随着切变率的升高逐渐上调 (182 .9± 2 8.2 ,2 2 9.3± 16.7,2 62 .6± 10 .5 ,P <0 .0 5 )。CD18和CD62L的表达与 [Ca2 + ] i变化呈显著线性相关 (r =0 .947,r =-0 .93 9,P <0 .0 5 )。结论 AP早期胰腺微循环紊乱与中性粒细胞激活密切相关的细胞内 [Ca2 + ] i的增加 ,在白细胞与内皮粘附过程中起重要作用。Objective To investigate the changes of cytosolic [Ca 2+ ]i and the correlation between expression of adhesion molecules (CD18,CD62L) and cytosolic [Ca 2+ ]i in neutrophil in pancreatic microcirculation of caerulein induced experimental acute pancreatitis (AP).Methods The Wistar rats (n=21) were randomized into 3 groups:normal control,AP 2?h group and AP 4?h group.The model of AP was established by subcutaneous injection of caerulein.The changes of neutrophil cytosolic [Ca 2+ ]i induced by shear stress was determined by flow cytometry.Result With the shear rate changing from low to high,cytosolic [Ca 2+ ]i was decreased slightly first under low shear rate and then increased gradually to a high level in the 3 groups (182.9±28.2,229.3±16.7,262.6±10.5 respectively,P< 0.05 ) as compared with stationary control.The positive correlation was found between the [Ca 2+ ]i and the expression of CD18 (r=0.947,P<0.05).The negative correlation was found between the [Ca 2+ ]i and the expression of CD62L (r=-0.939,P<0.05).Conclusion The increase of cytosolic [Ca 2+ ]i is highly related to the activation of neutrophil,which may play an important role in neutrophil adhesion to endothelium in the early phase of acute pancreatitis.
