摘  要:目的 了解儿童骨髓基质细胞的起源、组成及传代过程中的细胞骨架。方法 采用体外培养和免疫细胞化学染色技术 ,观察波形纤维蛋白 (Vimentin)、结蛋白 (Desmin)、α -平滑肌肌动蛋白 (α -SmoothMuscleActin)和VIII因子相关抗原 (VonWillebrandFactor)在儿童骨髓基质细胞系中的表达。结果 Vimentin在原贴壁基质细胞系由原代传至 5代的细胞中 ,其表达逐渐下降 ,而在再贴壁基质细胞传至第 3代中表达最高 ;Desmin、α -SmoothMuscleActin单克隆抗体和VonWillebrandFactor抗体染色呈阴性。结论 本结果提示本文中骨髓基质细胞来源于间充质 ,Vimentin的表达与细胞的分化、增殖有关 ,体外培养时骨髓基质细胞的组成、分化可能与培养条件有关。Objective To elucidate the origine,components of child bone marrow stromal cells and the variation of cytoskeleton during the process of repopulation.Methods To invistigate the expression of Vimentin, Desmin, α-Smooth muscle actin and Von willebrand factor in the Child marrow stromal cell line by in vitro culture and immunocytochemistry staining.Results The expression of Vimentin declined gradually during passaging process from the primary generation to the fifth generation, while the highest level of Vimentin expression was obtained on the third generation cells derived from the secondary adherent cells, however the other stainings were negative for the monoclonal antibodies against Desmin, α-Smooth muscle actin and Von willebrand factor.Conclusions The results indicates that the marrow stromal cells originate from mesenchyma.Additionaly the expression of Vimentin correlates with the differentiation and proliferation of stromal cells. The compenent and differentiation of marrow stromal cells cultured in Vitro may be related to culturing condition.
