摘 要:目的:采用高效液相色谱(HPLC)法测定硝苯地平临床治疗血药浓度。方法:选择乙酸乙酯-正己烷(3:2)混合液为提取溶剂,在Hypersil-BDS(250 mm×4.0 mm,ID 5 μm)分离柱上,以乙腈-20 mmol/L。磷酸二氢钠(60:40)为流动相,紫外吸收波长350nm检测临床样本。结果:硝苯地平血浆样品在5.0~200.0 ng/ml范围内线性关系良好(r=0.999 9,n=5),回收率>95.0%,日内日间的RSD<8.0%。结论:本法简便、准确、重现性好,适合于硝苯地平临床治疗血药浓度的定量监测。Objective: To monitor the clinical therapeutic concentrations of nifedipine by HPLC method. Methods: Plasma nifedipine was extracted using the mixture of ethyl acetate-n-hexane (3:2). The samples were analysed on a Hypersil-BDS(250 mm × 4. 0 mm,ID 5 μm)with acetonitrile-20 mmol/L sodium dihydrogen phosphate(60:40)as mobile phase. The detection wavelength was 350 nm. Results: A good linearity within the range of 5. 0-200. 0 ng/ml(r=0. 999 9) for nifedipine was obtained. The recovery rate was higher than 95. 0%. The inter-day and intra-day RSD were less than 8.0%. Conclusion: This method is simple, accurate and reproducible, and suitable for the determination of clinical therapeutic concentrations of nifedipine.
