摘 要:目的 观察葛根素在大鼠脑复苏时对海马CA1区神经细胞凋亡相关基因的影响。方法 将实验大鼠随机分为空白对照组、缺血再灌注组、葛根素组 ,采用Pulsinelli4VO法 ,制备大鼠全脑缺血再灌注模型 ,利用免疫组化法、原位末端标记法检测大鼠脑缺血再灌注不同时间内Fas、p5 3蛋白表达的水平及凋亡细胞数的变化。结果 ①脑缺血 10min再灌注 6h ,海马CA1区有少量p5 3蛋白表达的阳性细胞 ,再灌注 12h后增多。②脑缺血 10min再灌注 3h ,Fas蛋白表达的阳性细胞增多 ;再灌注12h ,Fas蛋白表达的阳性细胞达高峰 ,后下降。③神经细胞凋亡数随再灌注时间延长而增加。④给予葛根素治疗后 ,上述变化均减轻。结论 葛根素脑复苏的作用机制之一可能是抑制或延迟脑缺血时细胞凋亡、调控基因Fas、p5Objective To observe the effects of puerarin on the expression of p53 pritein and Fas protein which relate to neuron apoptosis after cerebral resuscitation Methods The rats were randomly divided into three groups:control group(only exposed anatomically without cerebral ischemia and reperfusion);normal saline(NS)group(global cerebral ischemia and reperfusion with administration of normal saline through abdomen);puerarin group(global cerebral ischemia and reperfusion with administration of puerarin through abdomen) Global cerebral ischemia and reperfusion was induced by means of pulsinelli's four blood vessel occlusion model After cerebral resuscitation,changes in protein expression of p53 and Fas were detected by immunohistochemiscal method.The number of neuronal apoptosis was assessed by TUNEL,the effects of puerarin intervention were observed Result ①Few expression of p53 positive cells were detected in hippocampal CA1 region at 6 h reperfusion after 10 min global cerebral ischemia and increased markedly 12?24?48 hours after the reperfusion ②Expression of Fas positive cell were increased in hippocampal CA1 region at 3 h and reached the peak level at 12 h reperfusion and then decreased gradually ③The number of neuronal apoptosis after cerebral ischemia reperfusion was increased ④In puerarin group,the expression of Fas and p53 were down-regulated The number of neuronal apoptosis was decreased Conclusion Puerarin could protect the brain from ischemia-reperfusion injury,the mechanism was related to its inhibiting or blocking apoptosis For cerebral resuscitation,treatment with puerarin can inhibit or delay apoptosis
