摘 要:目的 探讨创伤后免疫功能紊乱状态与巨噬细胞亚群的关系。方法 采用创伤失血模型 ,运用流式细胞仪分析单克隆抗体ED1、ED2单克隆抗体双标记表达情况 ,测定腹腔巨噬细胞 (pM)培养上清白细胞介素 6 (IL 6 )、肿瘤坏死因子 (TNFα)含量 (ELISA法 )。结果 正常大鼠pM分为ED1+ ED2 -亚群、ED1+ED2 + 亚群 ,无单纯的ED2 + 巨噬细胞亚群 ,创伤后ED1+ ED2 -亚群比例增加 ,ED1+ ED2 + 亚群比例降低 ,同时TNFα、IL 6分泌增加 ,内毒素 (LPS)刺激可增加TNFα、IL 6分泌水平。结论 创伤后细胞因子分泌紊乱可能与ED1+ ED2Objective To discuss the relationship between functional disorder of the immunological state and macrophage sub populations following trauma and hemorrhage.?Methods The expression condition of ED1?ED2 monoclonal antibody on peritoneal macrophage was assayed by flow cytometry and IL 6?TNFα content of peritoneal macrophage secretion was determined by ELISA method at various time points following trauma hemorrhage.?Results Peritoneal macrophage can be divided into ED1 + ED2 - macrophage sub population,ED1 + ED2 + macrophage sub population,but lack of pure ED2 + macrophage sub population;ED1 + ED2 - macrophage sub population increased and ED1 + ED2 + macrophage subpopulation decreased following trauma,content of secretion of TNFα and IL 6 increased after trauma was enhanced by stimulation with LPS.?Conclusion Cytokine parasecretion following trauma hemorrhage probably correlates with the differentiation and transformation of ED1 + ED2 - macrophage sub populations.
