摘 要:目的 探讨大鼠血清孕激素 (P)水平对胚胎“植入窗”的预测作用。方法 收集自然交配孕 3~ 10天大鼠的血清及子宫组织 ,以动情期雌鼠为对照。通过组织学切片确定各孕天发生胚胎着床的大鼠数 ,以 P2 .5~P97.5间距表示胚胎“植入窗”。采用放射免疫法测定大鼠血清 P水平。用免疫组化 L s AB法和计算机图像分析检测孕激素受体 (PR)在孕早期大鼠子宫内膜及胚胎滋养层细胞中的表达。结果 胚胎着床的中位数时间为 6 .6 5天 ,大鼠胚胎“植入窗”在孕 4.6 5~ 8.6 8天之间。孕 3~ 10天大鼠血清 P曲线的峰值出现在“植入窗”内 (孕 5~ 8天 )。PR在大鼠内膜腺、腔上皮、基质细胞、蜕膜细胞及滋养层细胞中均有表达 ,其表达强度曲线的峰值也出现在大鼠胚胎的“植入窗”内 ,并与血清 P水平呈显著性正相关 ;PR在大鼠滋养层细胞中的表达随妊娠期增加而升高 ,与血清P无显著性相关。P、PR与胚胎植入时间均呈显著性相关 (相关系数分别为 0 .86 8和 0 .72 4,P<0 .0 5 )。结论 P通过上调自身受体 ,对胚胎“植入窗”具有重要调节作用 ,P可能是胚胎“植入窗”的启动因子。连续监测血清 P可作为确定“植入窗”的生化标记。Objective To evaluate the possibility of using progesterone as a predictable indicator on 'implantation window' in rat. Methods The uterus and serum of pregnant rat were collected during 3-10 days of gestation, and the rat in estrus was used as control.The number of rats with embryo nidation occurring per day of gestation was determined by histological inspection, and the implantation window was defined by the 2.5 th to 97 5 th percentile.The serum progesterone (P) concentration was measured by radioimmunoassay; the expression of progesterone receptor (PR) in endometrium and trophoblast cell was detected by histoimmunochemistry and quantified by computer image analysis. Results The implantation happened on the 6.5 th day of gestation (median); the implantation window lay between 4.65 and 8.68 days of gestation (2.5 th to 97.5 th percentile).The serum P concentration of pregnant rats varied like a wave and reached peak during the embryo 'implantation window' (5-8 days of pregnancy). PR mainly expressed in luminal and glandular epithelium, endometrial stromal cell and trophoblast cell; the intensity of its expression, showing significant correlation with serum P, appeared like wave, and its peak lay between implantation window too.The intensity of PR expression in trophoblast cell rose with pregnant date and showed no significant correlation with serum P. Both P and PR were significantly correlated with implantation time of embryo.Conclusion By up-regulating of its own receptor, P has some important roles in embryo implantation and may be the promotor of implantation window. Successive monitoring of P serum concentration can be used as a biochemical indicator of implantation window.
