摘 要:目的 对比研究加替沙星与左氧氟沙星体外抗菌活性和甲磺酸加替沙星血清杀菌效价。方法 采用琼脂平板稀释法测最低抑菌浓度(MIC),MIC以左氧氟沙星为对照药,对8名健康受试者以400 mg甲磺酸加替沙星静滴,采静滴后即刻及给药后24h的血,采用微量稀释法测定血清杀菌效价(SBT)。结果 对革兰阳性菌的体外抗菌活性,加替沙星明显强于左氧氟沙星,其MIC90较左氧氟沙星低2~4倍;对革兰阴性菌,加替沙星与左氧氟沙星的体外抗菌活性相似;加替沙星对金黄色葡萄球菌、表皮葡萄球菌、肺炎克雷伯菌、阴沟肠杆菌的峰时SBT≥1:8的百分率分别为100%、90%、100%、100%,对粪肠球菌、大肠埃希菌、铜绿假单胞菌的峰时SBT≥1:8的百分率为50%、60%、50%。结论 加替沙星对常见致病菌有较强的体内外杀菌活性,提示加替沙星为更新一代广谱、强效、长效的氟喹诺酮类药;对革兰阳性菌的抗菌活性比左氧氟沙星显著增强。OBJECTIVE To evaluate antibacterial activity in vivo and in vitro of gatifloxacin. METHODS Minimal inhibitory concentration (MIC) and serum bactericidal titer (SBT) were determined by agar disk dilution method and microplate method respectively. RESULTS MIC90 of gatifloxacin against G+ bacterium was twice to fourfold as that of levofloxacin. MIC of gatifloxacin against G- bacterium was same as levofloxacin. The percentage of peak SBT≥1 : 8 with gatifloxacin aagainst Staphylococcus aureus, S. epidermidis, Klebsiella pneumoniae, En-terobacter cloatae was 100%, 90%, 100% and 100%, respectively and against Enterococcus faecalis ,Escherichia coli, Pseudomonas aeruginosa was 50%, 60%, 50%, respectively. CONCLUSIONS The antibacterial activity in vivo and in vitro of gatifloxacin against common bacterium is strong, especially that against G + bacterium is stronger than other quinolones.
