摘  要:本研究以 3个高抗粗缩病的玉米自交系 (P138、齐 319、H2 1)和 3个感病玉米自交系 (478、 10 7、Mo17)以及杂交组合 P138× 4 78为材料 ,寻找与玉米粗缩病 (MRDV)基因紧密连锁的分子标记 ,并对其辅助选择效果进行了初步探讨。结果表明 :引物 S37所扩增的多态性片段 S37+ 2 0 0 0 和 S86所扩增的多态性片段 S86 - 1 30 0 与 MRDV抗性基因相连锁 ,S37+ 2 0 0 0 和 S86 - 1 30 0 分别是该基因的正负 RAPD标记带。利用正标记带 S37+ 2 0 0 0 和负标记带 S86 - 1 30 0 对 F2 群体和 F3株系连续进行两个世代的选择 ,选择效果明显。正标记随世代增加抗病率上升 ,F2 、 F3、 F4 群体的抗病株率分别为 87.75 %、91.2 0 %和 94 .79% ;负标记随世代增加抗病率下降 ,F2 、 F3、 F4 群体的抗病株率分别为 87.75 %、 85 .32 %和 75 .39%。Maize rough dwarf disease, caused by Maize rough dwarf virus (MRDV), is the most widespread and serious virus disease of Maize ( Zea mays L.) in China. The objectives of this study were to identify RAPD markers linked to MRDV resistant genes and to verify the effects of marker assisted selection using 3 resistant maize inbred lines (P138, H21, Qi319), 3 susceptible maize inbred lines (478, 107, Mo17) and a hybrid (P138×478) progenies in the experiment. The main results are as follows: The results of RAPD marker identification showed that the polymorphic DNA band (S37 +2000 ) amplified by S37 and band (S86 -1300 ) amplified by S86 were linked to MRDV resistant genes. That is to say, the bands S37 +2000 and S86 -1300 were respectively the positive and negative RAPD markers linked to MRDV resistant genes. Two generations of marker assisted selections were carried out continuously on F 2 population and F 3 families using positive and negative marker bands, S37 +2000 and S86 -1300 , and the effects were evident. In using the positive marker, the resistance of progenies was significantly improved. The percentages of resistant plants of F 2, F 3 and F 4 populations were respectively 87.75%, 91.20% and 94.79%. By selecting the negative marker, the resistance of progenies was dramatically decreased. The percentage of resistant plants of F 2, F 3 and F 4 were respectively 87.75%, 85.32% and 75.39%.
