摘 要:通过对合成体系各组分对产物形成的影响的研究 ,分别求得了底物邻苯二酚、丙酮酸及乙酸氨的表观米氏常数、表观底物抑制常数和最适底物浓度 ,并得出合成左旋多巴的最适反应体系为 :1%丙酮酸 ,1.2 %邻苯二酚 ,2 %乙酸铵 ,0 .1%EDTA ,0 .2 %亚硫酸钠 ,用氨水调 pH至 8.0。加入从与合成体系等体积培养液中收获的重组大肠杆菌细胞 ,于 15℃反应 16h ,L -DOPA的产量为 15 .36 g/L。The apparent Michaelis constant, the apparentsubstrate inhibition constant and the optimal substrate concentration for the substrate pyruvate, catechol and ammonium acetate were calculated respectively from the studies on the effect of the components of the L-DOPA synthesis system on the L-DOPA production. The best reaction system contains 10g/L pyruvate , 12g/L catechol, 20g/L ammonium acetate ,1g/L EDTA,2g/L sodium sulfite and the cells from the fermentation broth which the volume was the same as reaction system, the pH was adjusted to 8.0 by adding ammonia. The mixture was incubated at 15℃for 16 hours. The productivity of L-DOPA was 15.36g/L.
