摘 要:研究pH值对转基因鱼腥藻培养的影响,发现培养基初始pH以自然pH8 3为宜。间歇流加稀盐酸控制过程pH值为8 5和9 0,生长较未流加快,每天流加稀盐酸一次控制过程pH为8 5,15d生物量大于3g/L,可溶性蛋白为0 3g/l藻干重,TNF表达量超过22%,生物量比未流加增长12 41%,而TNF表达量未受影响。Effect of pH was studied on the culture of recombinant Anabaena sp.PCC7120.The results showed that the optimal initial pH was 8.3.When pH was controlled to 8.5~9.0 by adding diluted HCl,the cell growth could be increased and the TNF expression could not be influenced.Under the optimal cultrue condition(pH was kept 8.5 during culture process),the biomass can exceed 3g/L,dissolvable protein was about 0.3g per gram dry weight cell,TNF expression level was more than 22%,the biomass increased 12.41% compared with cultured under nature pH.
