摘  要:利用高等植物类受体蛋白激酶基因的保守域设计简并引物 ,通过RT PCR方法 ,从大豆叶片中克隆到两个新的、可能的类受体蛋白激酶基因的部分cD NA片段。对其基因结构的分析表明 :在RLPK2的激酶保守域Vib与IX之间有一个 40 7bp长的内含子。利用RT PCR方法对它们的表达特性进行初步研究 ,发现这两个基因可能参与了对大豆叶片衰老和Partial cDNA fragments of two putative receptor-like protein kinases, RLPK2 and RLPK4, were cloned from soybean leaves with/without senescence-inducing treatment by RT-PCR (Fig.1). Sequence alignments (Blastp) suggested they have significant homology with eukaryotic protein kinase domain. FASTA search revealed similarities both between RLPK2 cDNA fragment and Arabidopsis thaliana receptor-like serine/threonine kinase (RKF1) gene (73% identity), and between RLPK4 cDNA fragment and Oryza sativa clone GRLG4 receptor kinase gene (74.519% identity) (Fig.2). Preliminary structural analysis showed a 407 bp intron between the Vib and IX subdomain of RLPK2 (Fig.1). The expression levels of both putative kinase genes in soybean leaf were up-regulated by senescence-inducing treatment. Exogenous 6-BA pretreatment partially reversed the effect of ST on the mRNA accumulation of RLPK2 gene while no obviously effect on gene expression of RLPK4 was detected (Fig.3). The specific expression patterns of these two genes implied their involvement in the soybean leaf senescence process and perhaps also in the senescence-retarding effect of cytokinins.
