心脉龙注射液对大鼠心肌细胞内游离Ca^(2+)的影响

摘　　要：目的探讨心脉龙注射液对大鼠心肌细胞内游离Ca2+的影响。方法采用荧光分光光度法测定给予不同剂量的心脉龙注射液后大鼠心肌细胞内Ca2+的浓度。结果心脉龙注射液3个剂量组(0.19、0.38、0.76g·L-1)均能增加心肌细胞内Ca2+含量,依次为169.18±11.64、233.26±10.69、164.25±10.34nmol·L-1,与对照组(120.64±3.02nmol·L-1)相比有显著性差异(P<0.01)。40μmol·L-1的维拉帕米对0.38g·L-1的心脉龙注射液所致的心肌细胞内Ca2+增加有抑制作用(抑制率为20%),而对0.19、0.76g·L-1心脉龙注射液所致的心肌细胞内Ca2+增加无明显抑制作用。结论心脉龙注射液强心作用与升高心肌细胞内Ca2+有关。Objective:To observe the effect of Xinmailong injection (XI) on free calcium ( Ca2+) content in rat myocardial cells. Methods:Free Ca2+content in rats treated with various dosages of XI were measured by fluorospectrophotometry. Results:Xinmailong injection significantly increased free Ca2+content and the content was 169.18±11.64, 233.26±10.69 and 164.25±10.34 nmol·L-1 in 0.19 g·L-1 , 0.38 g·L-1 and 0.76 g·L-1 XI group respectively, the differences being significant as compared with 120.64±3.02 nmol·L-1 in the control group (P < 0.01). Verapamil ( 40 μmol·L-1 ) induced the increased Ca2+content in 0.38 g·L-1 XI group at the rate of 20 %while had no effect on that in 0.19 g·L-1 XI group and 0.76 g·L-1 XI group. Conclusion:The positive inotropic action of Xinmailong injection in rats is related to the increase of free Ca2+content in myocardial cells.
