摘 要:简化了的禾谷多粘菌(Polymyxa graminis Ledingham)休眠孢子提取方法为:洗净干燥麦根,经高速粉碎机粉碎,称1g浸泡于15ml 1%偏磷酸钠和0.25%吐温20的混合提取液中,4℃下过夜。在强度为1000KHz的超声波发生器上处理45min,过滤,在血球计数板上计0.03mm^3的体眠孢子数。此提取法省去了组织捣碎机捣碎和低速离心等程序,因此工效比原方法提高3倍以上。比较了简化与不简化2种方法提取不同大麦品种根部所得的孢子数量,探讨了样品放置在超声波发生器上不同部位对休眠孢子提取效果的影响。A simpoified procedure to isolate the resting spores of Polymyxa graminis Ledingham is described as follows: the diseased barley roots infected by P. graminis were washed and dried, then ground with high speed blender into powder. One gram of root powder was taken to soak in 15 ml solution of 1 % sodium metaphosphate mixed with Tween 20 and overnight at 4℃. This was followed by ultrasonic treatment at 1000KHz for 45 minutes.The suspension was filtrated and taken to observe under micrscope fot the resting spores. The number of resting spores per gram of root sample was calculated by counting those spores with haemacytometer using phase contrast microscopy at 400 magnification. The efficiencies of spore extraction affected by sample positions in ultrasonic machine was examined and the procedure to extract the amount of spores from different barley varieties was compared with the original method.
