摘  要:目的:研究黄酮类化合物木犀草素(luteolin,Lu)治疗肝纤维化的作用。方法:30只Wistar大鼠随机分成正常对照组,实验对照组和Lu实验组,实验对照组和Lu实验组给予8wk CCl_4和酒精饮料制备肝纤维化动物模型,Lu实验组从实验开始就用20mg.Kg^(-1).day^(-1)LU灌胃。实验结束后,进行病理学检测和测定肝组织羟脯氨酸含量,用Northern Blot检测Ⅰ型前胶原mRNA表达,并测定了肝组织MDA含量和基质金属蛋白酶(MMPs)的活性。结果:经Lu治疗后,肝脏纤维化程度降低,羟脯氨酸含量减少;同时观察到Lu使肝脏Ⅰ型前胶原mRNA表达降低(5.2±1.4 vs 3.8±1.1,P<0.05)和MDA含量显著减少(12.7±2.6 vs 8.2±2.0μmmol/g,P<0.05),但MMPs的活性没有改变。结论:Lu通过清除自由基,抑制胶原基因表达,在动物体内具有防治肝纤维化作用。AIM: To investigate the role of luteolin (Lu) in the treatment of liver fibrosis. METHODS: Thirty Wistar rats were randomly divided into three groups, including normal control group, experimental control group and Luteolin group. The liver fibrosis was induced by carbon tetrachloride and alcohol drink for a period of 8 weeks. Luteolin group was given 20 mg.kg^(-1).day^(-1) Lu by injecting into stomach from the beginning of experiment The liver fibrosis was determined by pathological examination and hydroxyproline contents in hepatic tissue. The type Ⅰ procollagen mRNA expression was further explored by Northern Blot. MDA contents and matrix metalloproteinases (MMPs) activities were also detected RESULTS: The Results showed that the liver fibrosis was improved according to pathological changes and hydroxyproline contents in liver tissue after Lu treatment. Type Ⅰ procollagen mRNA expression and MDA contents were significantly decreased respectively (5.2±1.4 vs 3.8±1.1,P<0.05 and 12.7±2.6 vs 8.2±2.0mmol/g, P<0.05), however, the MMPs activities were not changed. CONCLUSION: Luteolin plays a role in treatment of liver fibrosis by inhibiting procollagen mRNA expression and scavenging free radicals.
