摘  要:目的 研究从猪心中提取和纯化缩醛磷脂 ,并测定其纯度 ,以利于进一步研究其生物学作用。方法 取新鲜猪心切块 ,绞碎 ,制成匀浆后 ,再经一系列过程包括①总脂的提取 ;②通过硅酸柱层析和硅胶薄板层析从总脂提取物中分离出磷脂酰乙醇胺 (PE)、磷脂酰胆碱 (PC)、磷脂酰肌醇 (PI)、磷脂酰丝氨酸 (PS)和鞘磷脂 (SM ) ,并从含PE浓度最高的柱层析液中分离出PE ;③碱水解制备缩醛磷脂酰乙醇胺 (plas PE) ;④通过硅酸柱层析和硅胶薄板层析从碱水解物中进一步纯化和鉴定 plas PE ;⑤分别用Iodinedisappearance法和Bartlett法检测 plas PE制剂中plas PE的含量及无机磷的含量 ,并计算出其纯度。 结果 ①从总脂提取液中分离出下列几种磷脂并经标准品鉴定出为PE、PI+PS、PC和SM ,其Rf值分别为 0 .87、0 .75、0 .54和 0 .37;②从含PE浓度最高的柱层析液中分离出PE ,并经碱水解 ,制备出plas PE ,其在薄板层析板上的Rf值为 0 .81 ;③plas PE制剂中无机磷含量为 3 .1 61 μmol/ml,plas PE含量为 3 .0 0 6μmol/ml,经计算 plas PE的纯度为 95 .1 0 %。 结论 猪心是缩醛磷脂含量丰富的器官 。Objective\ To study the extraction and purification of plasmalogen from pork heart, and to estimate its degree of purity so as to further study the biological function of plasmalogen. Methods\ A port heart, after being chopped and ground, was made into thick liquid, so that ①total lipid was extracted; ② PE?PC?PI?PS and SM were separated from the extraction of the total lipid with silicic acid chronatography and thin layer chromotogram, and PE was also separated from the liquid of the column with thick concentration of PE;③ alkaline hydrolysis was prepared to get acetal PE; ④ acetal PE from the alkaline hydrolysis was further purified and identified with silicic acid chronatography and thin layer chromotogram ;⑤the concentration of acetal PE and inorganic phosphorus in the preparation of acetal PE and their degree of purity were all estimated with the method of Iodine disappearance and Barlett. Results\ ① Several kinds of phospholipids had been extracted from the lipid extraction and compared with standard phospholipid preparations. Those were PE, PI+PS, PC and SM, and their Rf were 0.87, 0.75, 0.54 and 0.37 respectively.② The PE was extracted from PE chromatography liquids with thick concentration and the PE plasmalogen was producted by alkaline hydrolysis. The PE plasmalogen's Rf is 0.81.③ The content of acetal PE in acetal preparation is 3.006 μmol/ml. The total inorganic phosphorous content acetal PE preparation is 3.16 μ1mol/ml, in addition, the degree of purity of acetal PE is 95.10%. Conclusion\ There are plentiful acetal phosphatide in fresh pork heart. It is valid procedure to produce acetal phosphatide with silicic acid chromatography and alkaline hydrolysis.talPEandtheirdegreeo
