摘 要:目的 比较 2月龄及 18月龄Wister大鼠阴茎海绵体组织中一氧化氮合成酶 (Nitricoxidesynthase ,NOS)的含量 ,讨论其生理意义并对阴茎勃起功能障碍发病的年龄依赖性作出相应的解释。方法 使用NADPH d脱氢酶组织化学染色方法观察并进行图像分析。结果 发现阴茎海绵体组织中NOS阳性成分是普遍存在的 ,特别是在阴茎动脉外膜层周围及血窦内皮细胞系统中的含量较多。 18月龄大鼠阴茎海绵体组织总体NOS染色水平下降 ,进一步行图像分析 ,测定阴茎海绵体组织中NOS的含量 ,结果显示 18月龄大鼠阴茎海绵体组织中NOS含量明显低于 2月龄大鼠。结论 阴茎海绵体组织中NOS含量下降是阴茎勃起功能障碍 (Erectiledysfunction ,ED)Objective To compare the distribution of nitric oxide synthase in rat penile tissue of 2 and 18 months.Methods Using NADPH-diaphorase histochemical method and imaging analysis,the distributions of NOS in penile erectile tissues of 2 and 18 month rats were investigated.Results The penile arteries were surrounded by NOS positive fibers,which were also seen in the trabecular meshwork of smooth muscle.Endothelial cells lining many blood vessels were stained,whereas in the cavernous spaces the endothelial cells exhibited darker staining.Compared with 2 month rats, the staining of penile tissue of 18 month rats was faint. The imaging analysis showed that penile tissue of 2 month rats contained more NOS-positive compositions than that of 18 month rats.Conclusions NO may be an important mediator of penile erection.Decreased NOS in penile tissue might be one of the reasons of age-dependent erectile dysfunction.
