摘 要:目的 建立血清中测定乙醇浓度的乙醇氧化酶法。方法 基于乙醇氧化酶氧化乙醇产生乙醛和H2 O2 ,偶联Trinder反应生成醌亚胺 ,可在 5 0 0nm波长比色测定 ,通过优化反应体系中各主要成分和测定条件 ,建立了本法。结果 本法的灵敏度 0 .5 g/L时 ,A50 0nm ,1cm>0 .0 8;线性范围 0 .12 5~ 5 .0 g/L ;批内CV为 1.0 2 %,批间CV为 2 .76 %;本法 (Y)与气相色谱法 (X)比较 ;Y =1.0 6X +0 .12 ,r =0 .989。结论 本法的灵敏度较高、线性范围宽、精密度和准确性均较好 ,适合临床实验室急诊测定用。Objective An automated enzymic method was estabilished for assay of ethanol in serum. Method The assay is based on the fact that ethanol is oxidated by oxidase to produce ethanol and H 2O 2 and red colour of quinoneimine dye is then formed by coupled Trinder′s reaction which can be read by colorimetric determination at 500 nm wavelength. The concentrations of main components in the reactive system and the conditions of analysis were estabilished by optimization. Reasult The analysis index of this method were as follows: The sensitivity was (ΔA 500nm,1cm)>0.08 for 0.5 g/L of ethanol; the linearity was 0.125~5.0 g/L; the precisions were 1.02 % for within-run CV and 2.76 % for between-run CV. Correlation between the reasults obtained by this method (Y) and gas chromatlographic method (X) was Y= 1.06X+0.12,r= 0.989. Conclusion This method had higher sensitivity, wide linearity, good precision and accuracy. It is suitable to use for the emergency test of clinical laboratories.
