摘  要:目的 探讨大鼠烧伤后肠粘膜上皮细胞凋亡情况及胰高血糖素样肽 2 (GLP 2 )对其的影响。方法  75只Wistar大鼠随机分为烧伤组、GLP 2治疗组与正常对照组。动物于伤后 6、1 2h、1、3、5d处死 ,检测肠粘膜上皮细胞凋亡数、Caspases 3酶活性及Bcl 2mRNA表达的变化。结果 烧伤后 6h~ 5d肠上皮凋亡细胞数较正常组明显增加 ,GLP 2治疗后在伤后 6、1 2h、1d、3d、5d均明显低于烧伤组 ;caspase 3的活性在伤后 1d明显升高 ,伤后 3d到峰值 ,然后迅速下降至正常范围内 ,GLP 2组caspase 3活性只在伤后 3d明显高于伤前 ,而与烧伤组相比则在伤后 1、3d活性明显降低。肠粘膜Bcl 2mRNA表达在伤后 1 2h开始低于正常 ,至伤后5d仍低于正常 ,GLP 2组Bcl 2基因表达在伤后 1、3、5d低于于正常组 ,但与烧伤组相比 ,GLP 2组在伤后 1 2h、1d、3d、5d高于烧伤组。结论 GLP 2可降低烧伤后肠粘膜上皮细胞凋亡 ,其机制可能与降低Caspases 3酶活性 ,促进BclObjective To explore the effect of glucagon like peptide 2 (GLP 2) on the apoptosis of intestinal epithelial cells in burn rat. Methods Wistar rats with 30% total body surface area Ⅲ degree burns were randomly divided into 3 groups: burn group, GLP 2 treated group and normal control group. Animals were killed 0, 6, and 12 h, and 1, 3, and 5 d after burn. The apoptosis of intestinal epithelial cells, activity of caspase 3, and expression of Bcl 2 mRNA were detected. Results The apoptosis of mucosal epithelial cells was significantly increased in rats of burn and GLP 2 treated groups, but the apoptosis of GLP 2 treated group was lower than burn group at 6 h,1,3,and 5 d postburn. The activity of caspase 3 was increased on 1 d and reached the peak on 3 d postburn in burn group, and was lower in GLP 2 treated group than burn group on 1 and 3 d postburn. The expression of Bcl 2 mRNA was decreased in 12 h postburn in both groups, however GLP 2 treated group was higher than burn group in 12 h,and 1,3,and 5 d postburn. Conclusion GLP 2 may protect intestinal mucosa from apoptosis in burned rats. The mechanism of protection maybe due to the inhibiting of the activity of caspase 3 and promoting the expression of Bcl 2.
