摘 要:目的 :研究葛根素对急性肾缺血再灌注损伤的保护作用及可能机制。方法 :无创动脉夹夹闭双侧肾蒂制成急性缺血再灌注损伤大鼠模型 ,静脉注射葛根素注射液 (30 mg/ kg)。采用硫代巴比妥酸法测定丙二醛(MDA )含量 ,采用亚硝酸盐法测定超氧化物歧化酶 (SOD)活性 ,放射免疫法测定血浆血栓素 B2 (TXB2 )及6酮前列腺素 F1α(6 keto PGF1α)含量。结果 :急性肾缺血再灌注损伤后 ,葛根素组大鼠 MDA含量显著下降 ,SOD活性明显提高 ,肾组织病理改变明显轻于对照组 ;同时 ,葛根素组在缺血后 30分钟时的 TXB2 及6 keto PGF1α含量均明显升高 ,4小时时恢复至缺血前水平 ,再灌注 18小时时再次升高 ,其中对照组 TXB2升高更显著 (P<0 .0 5 ) ,而 6 keto PGF1α升高与对照组相比无显著差异。各生理参数变化 2组间无差异。结论 :葛根素对急性肾缺血再灌注损伤具有保护作用 ,其机制可能与其抗氧化损害的作用及改善缺血后血浆TXA2 PGI2 平衡失调有关。Objective:To investigate the role of puerarin on acute renal ischemiareperfusion injury and its mechanism.Methods:Acute renal ischemiareperfusion iniury rat model was established by placing the atraumatic vascular clamp in renal pedical for 30 minutes.Rats in puerarin group were given puerarin injection 30 mg/kg before ischemia.Malondialdehyde (MDA) assay for lipid peroxides in rats was detected by thiobarbituric acid reaction,superoxide dismutase (SOD) activity was detected by subnitrate method,and thromboxane B 2(TXB 2) and 6ketoprostaglandin F 1α (6ketoPGF 1α ) were detected by radioimmunoassay.Results:After renal ischemiareperfusion injury in rats,the level of MDA was lower and the activity of SOD was higher in puerarin group than those in the control.Much less necrotic tubular cells were found in puerarin group.At the same time,the level of TXB 2 increased significantly,but 6ketoPGF 1α didn't increase significantly after reperfusion 18 hours in puerarin group than those in the control.There was no difference of the vital signs change between 2 groups.Conclusions:Chinese herb puerarin can protect kidney against ischemia after acute renal ischemiareperfusion injury which may associate with the action of against oxidation and change the balance of plasma TXA 2PGI 2 in rats.
