摘 要:目的 :研究人参茎叶总皂苷 ( GSL)对体外培养的视网膜色素上皮细胞 ( RPE)增生的直接抑制作用及机制。方法 :通过活细胞计数法与 MTT比色法分别检测不同浓度 ( 4 0 0、30 0、2 0 0、1 0 0、5 0、1 0 mg· L-1)的 GSL和 GSL 30 0 mg· L-1在不同作用时间 ( 6、1 2、2 4、36、48、72、96和 1 2 0 h)对RPE细胞增生及代谢的影响。结果 :GSL组细胞增殖受抑并出现细胞脱落 ,GSL 40 0 mg· L-1具有最强的抑制效应 ,明显抑制效应在 6h出现 ,96h达高峰。 GSL组 A值明显低于对照组 ,RPE细胞生存率下降。结论 :GSL可能通过阻滞钙通道、干扰 RPE细胞代谢对 RPE细胞增殖具有剂量 -效应和时间Objective: To investigate the effect of Ginsenoside of stems and leaves (GSL) on proliferation of cultured retinal pigment epithelia and the possible mechanisms. Methods: Cultured RPE cell were treated with GSL by various concentrations(500,400,300,200,100,50,10 mg·L -1 ) and with GSL 300 mg·L -1 at different time intervals (6~120 h),cell proliferation and metabolism were evaluated by cell count method and MTT colorimetric assay. Results: There was inhibitory effect on proliferation of RPE cells and some cells detached from the wall in GSL group. The best concentration of GSL was 400 mg·L -1 . The significant inhibitory effect of GSL appeared at 6 hr, and the strongest inhibition of GSL occurred at 96 h. The A value in GSL groups was significantly lower than that of control group and the survival rate of RPE cells lowered. Conclusion: GSL may inhibit the proliferation of RPE cells in a dose dependent and time dependent manner, the probable mechanism was that GSL could block the calcium channel and disturb the metabolism of RPE cells.
