异基因造血干细胞移植后IL-2与急性移植物抗宿主病关系的研究

摘　　要：目的 :研究异基因造血干细胞移植 (allo- HSCT)后细胞因子 IL - 2的变化与急性移植物抗宿主病 (a GVHD)的关系 ,探讨 IL - 2在临床 a GVHD发病中的作用。方法 :30例恶性血液病患者行 allo- HSCT,采用双夹心 EL ISA法检测 a GVHD发生时 15例患者外周血单个核细胞分泌的 IL - 2浓度 ,采用逆转录聚合酶链反应 (RT- PCR)法检测 IL - 2的基因表达 ,并与未发生a GVHD患者进行比较。 结果 :所有患者均获得造血重建 ,15例发生 a GVHD(10例 度 a GVHD,5例 - 度 a GVHD) ,发生a GVHD患者的 IL - 2浓度以及基因表达阳性率均显著高于未发生 a GVHD者 ,并且 IL - 2浓度随着 a GVHD严重程度的增高而增高。 结论 :IL- 2在临床 a GVHD发生中起重要的正向调节作用。Objective: To investigate the relationship between interleukin 2 and human acute graft versus host disease (aGVHD). Methods: A total of 30 patients undergoing allogeneic hematopoietic stem cell transplantation (allo HSCT) were included in this study. IL 2 produced by peripheral blood mononuclear cells during aGVHD were measured using ELISA, IL 2 gene expression were analyzed using reverse transcriptase polymerase chain reaction (RT PCR), and the results were compared with those without aGVHD. Results: All patients achieved engraftment, 10 patients developed grade ⅠaGVHD, 5 developed grade Ⅲ Ⅳ aGVHD. Patients with aGVHD had significantly higher IL 2 levels than those without aGVHD. The maximum levels of IL 2 correlated significantly with the severity of aGVHD. Conclusion: IL 2 plays important roles in the development of human aGVHD.
