中药注射液喘可治对艾滋病猴模型Th17和Treg平衡的影响

摘　　要：【目的】研究温阳补肾中药注射液喘可治对艾滋病模型猴辅助性T细胞17/调节性T细胞(Th17/Treg)平衡的影响。【方法】选用12只猴免疫缺陷病毒(SIV)慢性感染的中国恒河猴,分别连续给予肌肉注射喘可治(治疗组,剂量为0.4 mg·kg-1·d-1)及生理盐水(对照组)2个月;于治疗前、治疗第4周和第8周分别检测SIV病毒载量,采用流式细胞仪分析外周血CD4、CD8、Th17(CD4+IL-17A+)和Treg(CD25+FoxP3+)细胞动态变化。【结果】喘可治对病毒载量,CD4、CD8细胞数无明显影响,可使CD25+FoxP3+和CD25+FoxP3-细胞显著减少(P<0.05),CD4+IL-17A+显著减少(P<0.05),Th17/Treg比值略降低。【结论】喘可治可同时降低SIV慢性感染猴外周血的Th17和Treg细胞水平。Objective To study the effect of Chuankezhi Injection （CI）, a Chinese herbal medicinal injection with the action of warming kidney-yang on the balance between T helper type 17 and regulatory T cells （Thl7/ Treg） in Chinese rhesus macaques infected with simian immunodefieieney virus（SIV）. Methods Twelve Chinese rhesus macaques infected with SIVmac239 were evenly randomized into two groups. Treatment group was given CI 0.4 mg ·kg^-1·d^-1 and control group was given normal saline by intramuscular injection every day for 2 consecutive months. The SIV load was detected before treatment, 4 and 8 weeks after treatment, and thedynamic changes of peripheral CD4 and CD8 T cells, Th17 （CD4＋IL-17A＋） and Treg （CD25＋FoxP3＋） were determined by flow cytometry. Results CI had no effect on SIV load and the count of CD4 and CD8 T cells, but had an effect on decreasing CD25＋FoxP3＋, CD25＋FoxP3-, CD4＋IL-17A＋ （P〈0.05）, and on reducing the ratio of Thl7fFreg slightly. Conclusion CI has an effect on decreasing Thl7 and Treg cells simultaneously in Chinese rhesus macaques with chronic SIV infection.
