摘  要:【目的】研究枳实白芍药对配伍高效液相色谱(HPLC)图谱中化学成分变化及与药效的相关性。【方法】采用冰醋酸灌肠与束缚夹尾相结合方法复制腹泻型肠易激综合征(D-IBS)大鼠模型,以小肠推进率、24 h粪点数和腹部回缩反射评分作为药效指标,并设计枳实白芍药对不同配伍组(质量比为1∶1、2∶1、1∶2),进行HPLC测定,将HPLC图谱中11个色谱峰与药效指标进行灰色关联度相关性分析。【结果】依据灰色关联度大小,确定各成分对小肠推进率贡献大小顺序:8>11>9>10>2>7>1>5>3;对24 h粪点数贡献大小顺序:8>9>5>11>7>10>1>2>3;对大鼠腹部回缩反射评分贡献大小:10>5>11>8>9>7>1>2>3。其中9号峰为柚皮苷、10号峰为橙皮苷、11号峰为新橙皮苷。【结论】枳实白芍药对可改善D-IBS大鼠胃肠道情况,与其所含的多种化学成分相关。Objective To study the changes of high performance liquid chromatograms of chemical components in herbal combination of Fructus Aurantii immaturus (Zhishi) and Radix Paeoniae (Baishao), and to explore the correlation of chemical components with pharmacological actions. Methods Diarrhea-predominant irritable bowel syndrome (D-IBS) rat model was established by enema with glacial acetic acid combined with body- constraining and tail-clamping stress method. Small intestinal propulsion rate, the number of feces granules within 24 hours and threshold value of abdominal withdrawal reflex were used as pharmacologic indexes. The chemical components of herbal combination of Zhishi-Baishao at mass fraction of 1:1, 2:1 and 1:2 were detected by high perfomance liquid chromatography, and then the correlation of 11 chromatographic peak values with pharmacologic indexes was investigated by grey relational analysis. Results Based on grey relational grades, the contribution of various components to small intestinal propulsion rate was shown as chromatographic peak 8〉11〉9〉10〉2〉7〉1〉 5〉3, the contribution to the number of 24-hour feces granules components shown as 8〉9〉5〉11〉7〉10〉1〉2〉3, and the contribution to threshold value of abdominal withdrawal reflex shown as 10〉5〉11〉8〉9〉7〉1〉2〉3. Of the chromatographic peaks, peak 9 was identified as naringin, peak 10 was identified as hesperidin, and peak 11 was identified as neohesperidin. Conclusion The improvement of herbal combination of Zhishi-Baishao on D-IBS rat gastrointestinal tract function is correlated with the multiple chemical components.
