扶正解毒方对肺腺癌A549细胞生长及凋亡的影响

摘　　要：目的:探讨扶正解毒方对肺腺癌A549细胞生长及凋亡的作用。方法:将10只大耳白兔随机分为扶正解毒方高剂量组、中剂量组、低剂量组,对照组,环磷酰胺(CTX)组5组,每组各2只。扶正解毒方高剂量组、中剂量组、低剂量组分别按16 g·kg-1、8 g·kg-1、4 g·kg-1剂量口服灌胃;对照组给予相同剂量生理盐水,每日1次,连灌1周;环磷酰胺(CTX)组第1天予20 mg·kg-1腹腔注射。末次用药后1 h,各组均在无菌条件下心脏采血,分离血清。采用血清药理学方法,对A549细胞株进行体外培养,然后分别加入扶正解毒方高剂量、中剂量、低剂量含药血清、对照组含药血清、CTX含药血清,通过光学显微镜直观法、细胞计数法、MTT法、流式细胞仪法等对A549细胞生长及凋亡指标进行测定。结果:A549细胞在显微镜下呈现典型的凋亡形态学改变;细胞生长曲线提示,扶正解毒方对A549细胞的抑制作用呈明显时效和量效关系;MMT法提示,扶正解毒方对A549细胞生长的抑制率随浓度的增加而增加,呈效应-剂量依赖关系;流式细胞仪检测显示,细胞凋亡率随着药物剂量的增加和作用时间的延长而增加,药物作用72 h后,扶正解毒方低剂量组、中剂量组、高剂量组凋亡率分别为(6.30±1.01)%、(15.72±1.54)%、(23.40±1.34)%,与对照组比较,差异均有统计学意义(P<0.05)。结论:扶正解毒方可抑制肺腺癌A549细胞生长、诱导细胞凋亡。Objective： To explore the effect of Fuzheng Jiedu Recipe on lung adenocarcinoma A549 cell growth and apoptosis. Methods：10 big ear rabbits were randomly divided into the Fuzheng Jiedu Recipe high dose group,middle dose group,low dose group and control group,cyclophosphamide（CTX） group,2 in each group. Fuzheng Jiedu Recipe high dose group,middle dose group,low dose group were respectively given oral lavage according to 16 g·kg- 1,8 g·kg- 1,4 g·kg- 1dose. The control group was given the same dose of normal saline,1 times a day,1 week even irrigation. The cyclophosphamide（CTX） group was given 20 mg·kg- 1intraperitoneal injection.Each group was under aseptic conditions heart blood and serum separation after 1 h at the end medication. By using the method of serum pharmacology A549 cell line in vitro,and then add the Fuzheng Jiedu Recipe high dose,medium dose,low dose the drug-containing serum medicated serum. Determine the A549 cell growth and apoptosis index by optical microscope,cell counting method,MTT method and flow cytometry instrument method. Results： The A549 cells present typical apoptosis morphological changes under a microscope. On the cell growth curve,the inhibitory effect of Fuzheng Jiedu Recipe for A549 cell was obvious limitation and concentration-response relationship. MMT method suggested that the inhibitory effect of A549 cell increased with the increase of the concentration,the effect of dose-response relationship. Flow cytometry instrument detection showed that apoptosis rate increased with the increase of drug dose and duration increased with the extension of drug effect after 72 h. The apoptosis rate of low dose group,middle dose group and high dose group was（6. 30 ± 1. 01） %,（15. 72 ± 1. 54） % and（23. 40 ± 1. 34） %. Compared with the control group,the difference had statistical significance（P 0. 05）. Conclusion： Fuzheng Jiedu Recipe can inhibit the growth of lung adenocarcinoma A549 cells and induce cell apoptosis.
