辛伐他汀对人牙髓细胞和脂多糖诱导的人牙髓细胞血管内皮生长因子的表达影响

摘　　要：目的:观察辛伐他汀对人牙髓细胞和脂多糖诱导的人牙髓细胞血管内皮生长因子表达情况。方法:用1×10-2μmol·ml-1辛伐他汀干预健康人牙髓细胞和10μg·ml-1脂多糖诱导的人牙髓细胞,分为健康对照组、辛伐他汀组、脂多糖组、辛伐他汀+脂多糖组,采用实时定量PCR和酶联反应吸附测定方法检测血管内皮生长因子的表达和分泌情况。结果:基因水平和蛋白水平,随着时间延长辛伐他汀与对照组相比促进人健康牙髓细胞血管内皮生长因子的表达和分泌(P<0.05);辛伐他汀对脂多糖诱导的人牙髓细胞表达和分泌血管内皮生长因子抑制作用不明显(P>0.05)。结论:1×10-2μmol·ml-1辛伐他汀能显著促进健康牙髓细胞分泌和表达血管内皮生长因子,有限抑制脂多糖诱导的牙髓细胞分泌和表达血管内皮生长因子。OBJECTIVE To observe the effects of simvastatin on human dental pulp cells（hDPCs） and the expression of vas- cular endothelial growth factor（VEGF） of hDPCs induced with lipopolysaccharides（LPS）. METHODS HDPCs and LPS-in- duced hDPCs were incubated with simvastatin, which were divided into the healthy control group, simvastatin group, LPS group and simvastatin ＋ LPS group. The changes of vascular endothelial growth factor （VEGF） were examined by real-time PCR and enzyme-linked immuno sorbent assay（ELISA）. RESULTS With the prolongation of time, simvastatin significantly enhanced hDPCs’ VEGF genes and protein levels（P〈0. 05）. But simvastatin had no significant inhibitory effect on the genes and protein levels of LPS-induced hDPCs （P〈0. 05）. CONCLUSION 1 × 10-2μmol·mL-1 simvastatin significantly increased VEGF protein and mRNA levels of hDPCs, but limitedly inhibited that of LPS-induced hDPCs.
