摘 要:目的:观察喜树果超微粉的显微特征并测定喜树碱溶出量。方法:气流粉碎制备喜树果超微粉和残渣,显微镜下观察其细胞学特征。在370/430 nm(λex/λem)波长下测定喜树果残渣和超微粉乙醇提取液中CPT含量。结果:镜下残渣细胞结构清晰可见,而超微粉无完整细胞存在,且粒径小于50μm。一定浓度的10-羟基喜树碱能猝灭喜树碱的荧光。喜树果超微粉的乙醇提取液中喜树碱含量有显著性差异(t=4.809,P<0.05)。结论:超微粉中活性成分溶出率高,荧光法测定喜树果药材中喜树碱含量。OBJECTIVE To observe the microscopic characteristics and determine the camptothecin contents of Fructus Camptothecae ultrafine powder. METHODS Fructus Camptothecae were shattered by jet mill and divided to ultrafine and fine powder. Cytological features were visualized using a BXS1 microscope. Using ethanol as solvent, the extracting solution of ultrafine and fine powder were measured fluorescence intensity at λex/λem (370/430 nm) to determine the content of CPT. RESULTS Cellular structure of fine powder was visible while ultrafine powder with a diameter under 50/~m had no complete cells. The fluorescence intensity of CPT was quenched by 10-hydroxy camptothecin in high concentrations, but it did not be interfered by a low concentration of 10-hydroxy camptothecin. The concentration of CPT between ultrafine and fine powder had a statistically significant difference(t = 4. 559,P〈0. 05). CONCLUSION Micronization can improve dissolution content of CPT. Fluo- rescence method can be used for quality evaluation of CPT in Fructus Camptothecae.
