摘  要:目的探讨莴苣苷B对大鼠脑缺血再灌注损伤时大脑皮质葡萄糖调节蛋白78(Grp78)和Caspase-3 mRNA表达水平的影响。方法用线栓法建立大鼠大脑中动脉缺血再灌注损伤模型,腹腔注射不同剂量的莴苣苷B。采用RTPCR技术检测大脑皮质中Grp78 mRNA和Caspase-3 mRNA的表达情况。结果再灌注后24 h和72 h检测表明,莴苣苷B组的Grp78 mRNA的表达与模型组相比均增加(P<0.05),总体上低、中剂量组作用明显,优于阳性对照药尼莫地平组(P<0.05);莴苣苷B组的Caspase-3 mRNA的表达与模型组相比均降低(P<0.05),总体上中、高剂量组作用明显,优于尼莫地平组(P<0.05)。结论莴苣苷B的抗脑缺血再灌注损伤作用在一定时间范围内与其持续上调皮质的Grp78 mRNA和下调Caspase-3 mRNA表达水平有关。AIM To investigate the influences of lactuside B on the mRNA expression of Grp78 and Caspase-3 in rats' cerebral cortex after cerebral ischemia injury. METHODS First,middle cerebral artery ischemia-reperfusion injury model was established. Each group was then treated with its corresponding medicine. Animals were separately sacrificed at 24 h and 72 h and the mRNA levels of Grp78 and Caspase-3 in rats' cerebral cortex were checked by RT-PCR. RESULTS Compared with the model group,all doses lactuside B treatment can increase the expression of Grp78 mRNA in rats' cerebral cortex. Moreover,the expression levels of Grp78 mRNA were higher in medium and low dose groups than those in the model group( P 〈 0. 05). Generally,the effects of low and medium groups were better than those of the positive control medicine and Nimodipine group( P〈 0. 05). Compared with the model group,receving lactuside B treatment could degrade the expression level of Caspase-3 mRNA( P 〈0. 05). Furthermore,the effects of medium and high dose groups were better than that of Nimodipine group( P〈 0. 05). CONCLUSION Lactuside B can play a positive role in resisting cerebral ischemia by increasing the expression of Grp78 mRNA and decreasing the expression of Caspase-3 mRNA.
