摘  要:家蚕质型多角体病毒(BmCPV)感染家蚕幼虫中肠上皮细胞而引起中肠型脓病,对病毒致病机理的研究有助于制定科学的预防策略。细胞自噬作为机体的一种免疫机制在抵抗微生物的感染中发挥作用。利用免疫组织化学法,检测BmCPV感染家蚕4龄幼虫中肠组织中的细胞自噬相关蛋白ATG8(autophagy-related gene 8)的表达变化:家蚕幼虫在感染BmCPV1-8 h内,中肠ATG8蛋白的表达显著高于正常水平;感染8-35 h,中肠ATP8蛋白的表达下降至正常水平;感染51 h,该蛋白质在中肠的表达又显著升高,至99 h达到高峰,且感染后62 h可以观察到中肠上皮层柱状细胞内有病毒多角体的产生。研究结果显示BmCPV感染家蚕幼虫的过程与中肠细胞自噬存在一定的关系。Bombyx moil cytoplasmic polyhedrovirus (BmCPV) infects epithelial cells of silkworm larval midgut, leading to midgut polyhedrosis. Understanding the mechanism of BmCPV infection can contribute to formulate scientific preven- tion strategies. As an immune mechanism, autophagy plays important role in the resistance of microorganisms. The ex- pressional variation of autophagy-related gene 8 (ATG8) protein in silkworm larval midgut of the 4th instar was assayed using immunohistochemical method. The results showed that the expressional level of ATG8 protein in midgut was sig- nificantly higher than the normal level at 1 h to 8 h after the silkworm larvae were infected by BmCPV, but decreased to background level at 8 h to 35 h post infection. Then the expressional level of ATG8 protein increased sharply again at 51 h and achieved a peak at 99 h. At 62 h post infection, viral polyhedra emerged in the columnar cells of midgut epi- thelial layer. These results suggest that there is certain relationship between autophagy response and BmCPV infection in silkworm larvae.
