红细胞保存液梯度添加法在不足量血制备去白悬浮红细胞中的应用

摘　　要：目的探讨红细胞保存液(MAP)梯度添加法在不足量血制备去白悬浮红细胞中的应用效果,提高不足量血制备去白悬浮红细胞的质量。方法测定去白浓缩红细胞和MAP的比重,通过比重换算去白浓缩红细胞重量梯度,根据重量梯度计算MAP的添加梯度,再计算添加MAP后的去白悬浮红细胞的相应重量梯度。留取不足量血,采用本站常规MAP添加法和MAP梯度添加法制备去白悬浮红细胞,分别测定血比容、血红蛋白含量、白细胞残留量、储存期末溶血率。结果 MAP梯度添加法制备的去白悬浮红细胞制品的各项质控检测指标符合国家标准要求,合格率高于MAP常规添加法,且MAP梯度添加法并不影响其他质控检测项目的制备质量。结论 MAP梯度添加法可以代替MAP常规添加法进行不足量血的制备。Objective To explore the effects of red blood cell（RBC）additive solution gradients on Leukocytes removement in small volume of blood preparation.and to improve the RBC quality in blood preparation.Methods Measure the RBC density and MAP were measured the weight gradient was converted according to the density,based on which the additive gradient of MAP was calculated.The inadequate volume of blood was prepared and both normal MAP additive process and MAP gradient addition method were used to prepare leukocytes-free RBC in additive solution;hematocrit,hemoglobin,white blood cell count residual and hemolysis ratio at the end of storage life were measured.Results The quality control test index of leukocytes-free RBC in additive solution prepared by MAP gradient addition method met the national standards and has a higher percentage of pass than the normal MAP additive process.In addition,MAP gradient addition method did not affect the preparation quality of other quality control items.Conclusion MAP gradient addition method can take place of normal MAP method to prepare the small volume of blood.
