摘  要:目的:优选降脂灵片的微波真空干燥工艺。方法:以浸膏相对密度、微波功率、干燥时间为自变量,大黄素、熊果酸、齐墩果酸提取量的总评"归一值"为因变量,通过Box-Behnken设计-响应面试验优化降脂灵片的微波干燥工艺。采用HPLC测定大黄素、熊果酸和齐墩果酸含量,流动相依次为甲醇-0.1%磷酸(80∶20),乙腈-甲醇-0.5%乙酸铵(67∶12∶21),检测波长分别为254,210 nm。结果:最佳微波真空干燥工艺为将浸膏浓缩至相对密度1.27 g·mL-1,微波功率511 W,干燥时间27 min;大黄素、熊果酸、齐墩果酸提取量分别为0.321,0.548,0.118 mg·g-1,与三者的预测值0.324,0.545,0.121 mg·g-1非常接近。结论:优选的干燥工艺稳定可靠,与热风干燥工艺相比指标成分含量无显著性差异,值得在中药物料的干燥工序中推广应用。Objective: To optimize microwave vacuum drying process of Jiangzhiling tablets. Method:Considering overall desirability of contents of emodin, ursolic acid and oleanolic acid as dependent variable, Box- Behnken design-response surface method was used to optimize microwave vacuum drying process of Jiangzhiling tablets with relative density of extract, microwave power and drying time as independent variables. HPLC was adopted to determine contents of emodin, ursolic acid and oleanolic acid with mobile phases of methanol-0. 1% phosphoric acid (80: 20) and acetonitrile-methanol-0. 5% ammonium acetate (67: 12:21 ), detection wavelengths at 254 and 210 nm. Result: Optimum microwave vacuum drying process was as follows: concentrated extract to a relative density of 1.27 g ·mL^-1, microwave power of 511 W, drying time of 27 min; extracting amounts of emodin, ursotic acid and oleanolic acid were 0. 321, 0. 548, 0. 118 mg .g^-1^, which were close to predictive values of 0. 324, 0. 545, 0. 121 mg·g^-1, respectively. Conclusion: This optimized drying process was stable and reliable, which had no significant difference in contents of index components by hot air drying process, it was suitable for application in drying process of materials of traditional Chinese medicine.
