摘 要:目的:采用HPLC法测定不同来源、不同等级的52批黄芪饮片中黄芪甲苷含量,为制订黄芪饮片等级标准提供研究基础。方法:采用Aglient SB-C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈-水(32∶68)为流动相,流速1.0 mL·min-1,柱温30℃,蒸发光散射检测器,漂移管温度109℃,载气流速3.0 L·min-1。结果:52批黄芪饮片中,黄芪甲苷含量最高为内蒙古赤峰黄芪须根,0.28%;含量最低为甘肃陇西的一等黄芪,0.022%。46批黄芪饮片各级之间的黄芪甲苷的含量利用SPSS15.0统计学软件进行方差分析,P<0.05。结论:不同等级的黄芪饮片中黄芪甲苷含量与饮片直径大小具有一定的相关性,饮片直径越大,黄芪甲苷含量越低。Objective: To determine the content of astragaloside Ⅳ in 52 batches of Radix Astragali medicinal slices from different sources and grades by HPLC,in order to provide the basis for formulating the grade standard of medicinal slices of Radix Astragali. Methods: The analysis was carried out on an Aglient SB- C18( 4. 6 mm × 250 mm,5 μm) column at 30 ℃ with the mobile phase of acetonitrile- water( 32 ∶ 68) at a flow rate of 1. 0 mL·min- 1. The ELSD conditions were: drift tube temperature 109 ℃,nebulizing gas flow rate 3. 0 L·min- 1. Results:In the 52 batches of astragalus root samples,the highest astragaloside content 0. 28% was detected in the samples of astragalus root fibrous from Chifeng,Inner Mongolia. The lowest astragaloside Ⅳ content 0. 022% was found in the sample from Longxi,Gansu province. The astragaloside content in the 46 groups of astragalus root slices at all grades was subjected to variance analysis using SPSS 15. 0 statistical software,and the result was P 0. 05. Conclusion:The content of astragaloside Ⅳ of samples with different sources and grades showed a significant negative correlation with the size of medicinal slices. The content of astragaloside Ⅳ decreased with the increasing size.
