摘 要:目的:观察姜黄素对大鼠肝纤维化肝脏组织TGF-β1(transforming growthfactor-β1,TGF-β1)/smad信号通路的影响以及探讨其抗肝纤维化的作用机制。方法:30只雄性SD大鼠随机分为正常组、肝纤维化组和姜黄素治疗组。肝纤维化组和姜黄素治疗组均使用将使用二甲基亚硝胺建立肝纤维化模型,姜黄素治疗组在造模同时给予姜黄素治疗。动物处死后,血标本检测肝功能,取肝脏组织,肝组织HE染色行纤维化分级,逆转录-实时荧光定量PCR(Reverse Transcription-Real time quantitative Polymerase Chain Reaction,RT-qPCR)检测肝脏TGF-β1、smad2、smad3、smad4、smad7的mRNA表达,蛋白质印迹法(western blot)检测肝脏TGF-β1、smad4、smad7的蛋白表达。结果:与模型组比较,姜黄素治疗组肝纤维化病理分级明显改善,大鼠肝脏组织TGF-β1、smad4的mRNA表达下降,smad7的mRNA表达升高,smad2、smad3的mRNA表达无明显改变;与模型组比较,姜黄素治疗组TGF-β1、smad4的蛋白表达下降,smad7的蛋白表达升高。结论:姜黄素在阻断大鼠肝纤维化发挥积极的作用,通过多位点影响肝组织TGF-β1/Smad通道可能是其阻止肝纤维化进程的机制。Objective:To observe how curcumin affects TGF-β1/Smad signaling pathway on liver tissue of rat liver and to explore its mechanism of anti-hepatic fibrosis.Methods:30 male Sprague-Dawley rats were divided into three groups(normal group,hepatic fibrosis group,curcumin treatment group) randomly.The animal model with hepatic fibrosis was established by injecting solution DMN(dimethylnitrosamine) into abdominal cavity in hepatic fibrosis group and curcumin treatment group.And rats in curcumin treatment group were given curcumin treatment while modeled.After all rats were killed,their liver function was detected,and hepatic tissue was observed by Hematoxylin-Eosin staining.Furthermore,RT-qPCR detected mRNA expressions of liver TGF-β1,Smad2,Smad3,Smad4 and Smad7,and western blot detected protein expressions of liver TGF-β1,Smad4 and Smad7.Results:Compared with the rats in hepatic fibrosis group,the degrees of fibrogenesis were much better,and mRNA expressions of TGF-β1 and Smad4 decreased,and mRNA expression of Smad7 increased in liver tissues of the rats treated by curcumin,but mRNA expression of Smad2 and Smad3 didn't change obviously.Compared with the rats in hepatic fibrosis group,protein expressions of liver TGF-β1and Smad4 decreased,and protein expression of Smad7 increased in liver tissues of the rats treated by curcumin.Conclusion:Curcumin played an active role in blocking rat liver fibrosis and it affected liver tissue TGF-β1/Smad channel by multiple ways,which might be its mechanism to prevent the progression of hepatic fibrosis.
