摘 要:目的:通过建立人胃癌SGC-7901/VCR皮下移植瘤模型,探讨温郁金逆转人胃癌多药耐药的作用及可能机制。方法:SPF级4~6周龄BALB/c裸小鼠50只,运用组织块法建立建立人胃癌SGC-7901/VCR皮下移植瘤模型,2~3周后选取瘤体大小相近裸鼠36只,随机分为6组(模型组,VCR组,温郁金低剂量组,温郁金高剂量组、温郁金低剂量联合VCR组及温郁金高剂量联合VCR组),2周后处死裸鼠,观察各组瘤体生长情况,计算抑瘤率,同时采用电镜观察瘤体超微结构改变。结果:人胃癌SGC-7901/VCR皮下移植瘤成瘤率100%,经过两周给药后,温郁金联合VCR干预组较之模型组比较瘤体明显缩小,其中以温郁金高剂量+VCR组体积最小,其抑瘤率为51.56%,与模型组及VCR组比较均有统计学意义(P〈0.05),瘤体超微结构发现温郁金联合VCR组核碎裂明显,密度明显升高,染色质染色加深,向边缘聚集,细胞核固缩变形呈不规则形,线粒体嵴部分缺失或溶解甚至消失呈空泡变性,部分可见典型的凋亡小体。结论:联合VCR组可明显抑制移植瘤的生长,提示温郁金可逆转SGC-7901/VCR皮下移植瘤对VCR的耐药性或增强VCR的细胞毒性及增强VCR诱导胃癌多药耐药裸鼠移植瘤细胞凋亡有关。Objective:To the possible mechanism of the reversal of multidrug resistance of Curcuma wenyujin by establishing the model of human gastric carcinoma SGC-7901/VCR transplanted tumor.Methods:After 2 ~ 3 weeks,36 from 50 BALB/c nude mice with SGC-7901/VCR transplanted tumor by sSubcutaneous implantation of tumor tissue blocks were divided into 6 groups,including the model group,VCR group,Curcuma wenyujin low dose group,high dose group,low dose of Curcuma wenyujin plus VCR group,VCR group of high dose of Curcuma wenyujin.Nude mice were killed after 2 weeks for oobserving the tumor growth,inhibition rate and the changes of ultrastructure by electron microscopy of tumor.Results:We found that Curcuma wenyujin alone had no significant inhibitory effect on transplanted tumor,but Curcuma wenyujin combined with VCR can inhibit tumor growth.Curcuma wenyujin high dose plus VCR group had the strongest inhibitory rate with minimum volume and had statistical significance as compared with VCR group(P〈 0.05).The tumor inhibition rate was 51.56% and there was statistical significance compared with the model group and VCR group(P〈 0.05).With electron microscopy,we found that all groups except the model groups can induce chromatin margination.In Curcuma wenyujin plus VCR groups,chromatin fragmentation and margination were obvious,and with the increasing dose of Curcuma wenyujin,the chromatin margination was obvious to a higher degree.Conclusion:Curcuma wenyujin combined with VCR can significantly inhibit the tumor growth,reverse the drug resistance of SGC-7901/VCR subcutaneous transplantation tumor of VCR or enhance the cytotoxicity of VCR and enhance the effection of VCR to induce apoptosis of transplanted tumor of nude mice on multidrug resistance of gastric cancer.
