复方藤梨根制剂对小鼠CT26移植瘤生长及对MVD影响的研究

摘　　要：目的:通过建立Balb/c小鼠CT26人工种植模型,研究复方藤梨根制剂对肿瘤抑制效果及对MVD表达的影响。方法:①100只7周龄雌雄各半Balb/c小鼠随机分为5组,即空白对照组、荷瘤对照组、低剂量给药组、中剂量给药组、高剂量给药组。各组分别皮下接种造模。②造模后第2 d开始连续灌胃3周。给药组予低浓度0.5 g/mL、中浓度1 g/mL、高浓度2 g/mL复方藤梨根制剂灌胃,空白对照组、荷瘤对照组每天以生理盐水灌胃。③所有小鼠第22 d处死,观察各组的瘤重、抑瘤率,免疫组化检测各组小鼠肿瘤CD34标记的MVD。结果:FFTLG低、中、高剂量组抑瘤率分别为-7.05%、23.03%、32.91%;中、高剂量给药组的瘤重与低剂量组和荷瘤组有明显差异(P<0.05),中、高剂量给药组的MVD对比荷瘤对照组有显著性差异(P<0.05)。结论:复方藤梨根制剂能抑制肿瘤生长,减少CT26瘤体中MVD,具有抗肿瘤血管生成作用。Objective：To establish a model of artificial planting with CT26 in Balb/c rats for the study of inhibitory effect of Fufang Tengligen mixture on tumour and the influence of MVD.Methods：①100 7 weeks old rats（half of male and half of female） were randomly divided into five groups：blank control group,tumor-bearing group,low-dose drug group,medium dose drug group,high dose drug group.Each group was separately molding.②All rats were given continuous gavage after molding on the second day,lasting for 3 weeks.Drug group were treated with low concentration of 0.5g/mL,middle concentration of 1g/mL,high concentration of 2 g/mL.Blank control group and tumor-bearing group were treated with NS every day.③All rats were executed on the 22nd day,taking out the tumor and weighing and measuring the volume as well as calculating the anti-tumor rate,and then we made the pathological section and did the immunohistochemistry.The expression of MVD in the tumor was detected.Results：The tumor inhibition rates in low,middle and high dose groups were-7.05%,23.03%,32.91%.In the high and middle dosage group,the tumor weight had significant difference compared with that in low dose group and tumor-bearing group（P 〈 0.05）.In contrast,high and middle dose MVD groups and tumor-bearing control group had significant differences（P 〈 0.05）.Conclusion：Fufang Tengligen Mixture can inhibit tumor growth and reduce tumor of CT26 in MVD with anti-tumor angiogenesis.
