百会穴长留针法对慢性应激抑郁大鼠中枢5-HT和DA水平的影响

摘　　要：目的:观察百会穴长留针法对慢性应激抑郁模型大鼠的效应,并从神经生化角度探讨针灸抗抑郁的作用机理。方法:选用36只SD雄性大鼠随机分为空白对照组、模型对照组、百会穴长留针组和百会穴普通留针组。除空白组外,其余3组大鼠采用孤养结合慢性轻度不可预见性应激进行造模,共接受21 d不可预见的刺激,造模同时百会穴长留针组和普通留针组分别给予百会穴长留针和普通留针治疗。用高效液相电化学法(HPLCECD)测定实验后大鼠海马和下丘脑组织匀浆的单胺类神经递质5-羟色胺(5-HT)和多巴胺(DA)的水平。结果:实验后与空白组相比,模型对照组大鼠下丘脑DA含量明显下降(P<0.05)。与模型对照组相比,百会穴长留针组下丘脑5-HT和DA及海马5-HT水平显著性升高(P<0.05),百会穴普通留针组下丘脑DA水平显著升高(P<0.05);同期组间比较显示,百会穴长留针组较普通留针组各指标(P>0.05),但有升高趋势。结论:百会穴长留针法可提高海马及下丘脑5-HT和DA水平,其机制可能与调节脑内单胺类递质有关。Objective：To observe the effects of long-time-retaining needle at Baihui point on rats and to explore the therapeutical mechanism of anti-depression from the perspective of neurobiology.Methods：32 male SD rats were randomly divided into 4 groups.There were normal control group,model control group,long-time-retaining needle at Baihui group and common-time-retaining needle at Baihui group.The models except for the normal control group were set up by combining both chronic mild unexpected stimulation and solitary feeding.The rats accepted various stimulations for 21 days.At the same time,long-time-retaining needle at Baihui group and common-time-retaining needle at Baihui group were given long-time-retaining needle and common-time-retaining needle therapy.After treatment,the levels of 5-hydroxytryptamine（5-HT） and dopamine（DA） in hippocampus and hypothalamus in experimental rats were detected by HPLC-ECD.Results：After the experiment,compared with the normal control group,the level of DA in hippocampus on the model control group was reduced（P 〈 0.05）.In long-time-retaining needle at Baihui group,the levels of 5-HT and DA in hypothalamus and 5-HT in hippocampus were raised,meanwhile,in common-time-retaining needle at Baihui group,the level of DA in hypothalamus was significant raised（P〈 0.05）.Lastly,compared with common-time-retaining needle at Baihui group,long-time-retaining needle at Baihui group was better but there was no significant difference between them（P〉0.05）.Conclusion：The therapy of long-time-retaining needle at Baihui can raise the levels of 5-HT and DA in hypothalamus and 5-HT in hippocampus,and the therapeutical mechanism of anti-depression may be relevant to the modulation of neurotransmitter.
