摘 要:为探索副鸡禽杆菌生长特性,比较不同血清型菌株生长特性的差异,采用分光光度法和菌落计数法分别测定副鸡禽杆菌血清A型(221株)、血清B型(0222株)、血清C型(Modesto株)3个参考菌株的生长曲线,对两种方法测定的生长曲线进行比较。结果表明,两种方法绘制的生长曲线差异较大,对数生长期和稳定期菌液OD 600nm值与活菌数对数之间呈现一元二次多项式回归关系。另外,3个血清型菌株的生长稳定期都比较短暂,各型菌株达到生长高峰的时间和活菌数都有差异。该试验为副鸡禽杆菌生物学特性的比较研究及疫苗的研制奠定了基础。To explore and compare the growth changes and growth characteristics of different serotype of Avibacterium paragallinarum,growth curves of three reference strains of serotype A(221strain),B(0222strain)and C(Modesto strain)were determined by spectrophotometry and colony count methods under the same culture conditions.Then two methods were compared.Results showed that the growth curves made by two methods were different.OD 600 nm and the logarithm of colony count in logarithmic phases and stationary phase showed a quadratic polynomial regression.In addition,stationary phases of three serotypes were short and differences were observed on reaching the growth peak and viable colony counts in three serotypes.This experiment laid a foundation for the further comparative study on the biological characteristics and vaccine development of Avibacterium paragallinarum.
