摘 要:目的观察黄芪对慢性哮喘豚鼠模型气道重塑的影响及机制。方法 SPF级雄性豚鼠40只随机分成:对照组、模型组、黄芪注射液组、地塞米松组,每组10只。采取HE染色观察气道重塑,图像分析仪分析肺组织气道重塑情况,免疫组化方法测气道中TGF-β1蛋白的表达。结果与对照组相比,模型组气道壁结构发生改变,支气管管腔变窄,气道上皮细胞脱落,平滑肌增生/增殖肥大。经黄芪或地塞米松治疗后上述改变明显减轻。模型组支气管壁厚度、支气管壁平滑肌厚度、支气管壁平滑肌细胞核数量分别较对照组显著增加。模型组TGF-β1蛋白的积分光密度值(IOD)均较正常组明显增高,黄芪治疗后TGF-β1 IOD值显著下降。结论黄芪注射液能够抑制慢性哮喘模型的气道重塑,其机制可能通过抑制TGF-β1的表达有关。Objective To investigate the expression of TGF-β1 in the airway epithelial cell of asthmatic guinea pigs and study the effects of Huang Qi on airway remolding in asthma. Methods Forty adult male guinea pigs were randomly divided into control,asthmatic,arsenic Huang Qi and dexamethasone groups. HE staining and the image analysis were used to analyse the information of the airway remodeling. Immunohistochemistry was used to detect the expression of TGF-β1 in the lung tissue. Results Compared to control group,the model group showed that proliferation of airway smooth muscle,reconstruction of air passage,narrowing of bronchial lumen and swelling of airway epithelial cells were significantly milder after treatment with Huang Qi or dexamethasone. The airway wall and smooth muscle layer were significantly thicker in asthma group than that of control group,the number of smooth muscle cell nucleus increased significantly. The integrated optical density( IOD) value of TGF-β1 were statistically significantly higher than that in control which was derreased after treated with Huang Qi. Conclusions There was TGF-β1-overproduction in airway epithelial cells of asthmatic guinea-pigs,which may contribute to airway remolding in asthma. Astragus could decrease airway remolding by inhibiting the expression of TGF-β1.
