摘 要:目的:从血液流变学和红细胞膜组分的变化探讨少腹逐瘀汤改善气虚血瘀模型大鼠血瘀证的作用机理。方法:40只Wistar大鼠,随机分为空白组、气虚血瘀模型组、少腹逐瘀汤高剂量组(4.8g/kg)、低剂量组(1.2g/kg),每组10只。除空白组外,其他各组复制气虚血瘀模型,每日1次,连续14d。各组灌胃给药,每日1次,连续给药15d。取血前大鼠禁食24h,不禁水。取血测定血液流变学指标以及红细胞膜组分相关指标。结果:模型组造模后大鼠体质量显著下降(P<0.01),毛色状态等变差。与空白组比较,模型组大鼠全血黏度及血浆黏度均显著升高(P<0.01),红细胞变形指数显著降低(P<0.01),膜上Na+-K+-ATP酶活力、唾液酸含量及巯基含量均显著降低(P<0.05,P<0.01),丙二醛(MDA)含量和膜胆固醇含量均显著升高(P<0.05);少腹逐瘀汤能够显著降低全血黏度和血浆黏度(P<0.05,P<0.01),可显著升高Na+-K+-ATP酶活力、唾液酸含量以及巯基含量(P<0.05),显著降低MDA含量(P<0.05,P<0.01)。结论:少腹逐瘀汤可能是通过提高Na+-K+-ATP酶活力、唾液酸和巯基含量,而增加红细胞膜表面负电荷,增加抗氧化性,减轻自由基对红细胞攻击程度,改善红细胞的变形性,达到改善气虚血瘀模型大鼠血瘀状态。Objective: To investigate the mechanism of improving blood stagnancy due to qi deficiency by Shaofu Zhuyu Decoction(SFZYT), from the changes of hemorheology and the components of erythrocyte membrane. Methods: Forty Wistar rats were randomly divided into 4 groups, control group, model group, SFZYT high-dose group(4.8g/kg) and low-dose group(1.2g/kg), with 10 rats in each group. Qi deficiency and blood stasis rats model were established, except rats in control group. The related indexes of hemorheology and the components of erythrocyte membrane were determined on the 15th day. Result: The state of rats in model group became deteriorated and the weight of rats decreased signii cantly(P〈0.01). Compared with control group, the whole blood viscosity and plasma viscosity, the contents of MDA and membrane cholesterol in the model group were increased signii cantly(P〈0.05). Red blood cell deformability, contents of sialic acid, hydrosulphonyl and the active of Na+-K+-ATP and the contents of MDA and membrane cholesterol in model group were all decreased signii cantly(P〈0.05). SFZYT could decrease the whole blood viscosity, plasma viscosity, the contents of MDA and membrane cholesterol signii cantly(P〈0.05). It also could increase the content of sialic acid, hydrosulphonyl and the active and Na+-K+-ATP signii cantly(P〈0.05). Conclusion: SFZYT could improve the blood stasis condition of model rats through increasing the enzyme activity of Na+-K+-ATP, the content of sialic acid, hydrosulphonyl, the negative charge of erythrocyte membrane and inoxidizability, reducing the damage of free radical on red blood cell and improving deformability of the red blood cell.
