基于U0126阻断ERK信号通路的电针阿是穴对兔钝挫伤后肌肉再生影响的研究

摘　　要：目的:观察电针阿是穴对应用细胞外信号调节激酶(ERK)抑制剂U0126的兔腰肌钝挫伤后增殖细胞核抗原(PCNA)、碱性成纤维细胞生长因子(bFGF)以及ERK1/2表达的影响。方法:40只雄性新西兰大耳兔随机分为空白组、模型组、模型抑制剂组、电针组、电针抑制剂组,每组8只。建立兔腰肌钝挫伤模型,造模后1d检测血清磷酸肌酸激酶(CK)含量,14d后取腰肌行HE染色,PCNA、bFGF的免疫组化染色以及ERK1/2的蛋白印记表达。结果:电针组的肌纤维平均横截面积(CSA)、每肌纤维细胞核数(NPF)、PCNA表达以及ERK1/2的表达均高于模型组与电针抑制剂组(P<0.05,P<0.01);电针组bFGF的表达明显高于模型组(P<0.01)。结论:应用ERK抑制剂U0126可部分阻断电针阿是穴对兔腰肌损伤后肌卫星细胞增殖的促进作用,bFGF/ERK信号通路可能是电针阿是穴促肌肉再生的主要途径之一。Objective： To observe the expression of proliferating cell nuclear antigen（PCNA）, basicfibroblast grow factor（bFGF） and extracellular signal-regulated kinase（ERK1/2） after acupuncture ouch point of rabbits with muscle regeneration after contusion based on the ERK signal pathway inhibited by U0126. Methods： 40 male New Zealand rabbits were randomly divided into blank group（BG）, model group（MG）, model with inhibitor group（MI）, electro-acupuncture group（EA） and an electroacupuncture with inhibitor group（EI）, 8 rabbits in each group. 1 day after contusion model of lumbar muscles was established, CK level in serum was tested. After 14 days, HE staining, PCNA and bFGF immuno-histochemical staining were used to dye the lumbar muscle of each rabbit. Western blot was used to detect the expression of ERK1/2 protein. Results： Compared with MGand EI, the average cross-sectional area（CSA） and nuclei number per musclefiber（NPF）, PCNA and ERK1/2 expression were all higher in EA（P0.05, P0.01）. Compared with MG, the expression of bFGF was higher in EA（P0.01）. Conclusion： ERK inhibitor U0126 could partly inhibit the satellite cell proliferation with electro-acupuncture ouch point in rabbits with muscle regeneration after contusion. BFGF/ERK signal pathway might be one of the mainly pathways of promoting muscle regeneration when acupuncturing at ouch point with electro-acupuncture.
