摘 要:目的:观察复方菖蒲益智汤对脑缺血再灌注损伤小鼠白介素-1β(IL-1β)、转化生长因子-β1(TGF-β1)表达水平的影响,探讨复方菖蒲益智汤防治脑缺血再灌注损伤的可能机制。方法:采用阻断双侧颈总动脉法制备脑缺血再灌注模型。放射免法检测各组小鼠血清中IL-1β的表达水平,免疫组织化学法检测各组小鼠脑组织TGF-β1的表达水平。结果:与假手术组相比,模型组小鼠IL-1β、TGF-β1的表达水平明显升高(P<0.05);与模型组相比,各用药组小鼠血清中炎症因子IL-1β表达水平明显降低(P<0.05),脑组织TGF-β1的表达水平明显升高(P<0.05),而复方菖蒲益智汤高剂量组尤为明显(P<0.05)。结论:复方菖蒲益智汤可能通过调节炎症因子的释放,抑制炎症反应,降低脑缺血再灌注损伤的程度。Objective: To observe the inl uence of Fufang Changpu Yizhi Decoction on the level of IL-1β and TGF-β1 in mice of cerebral ischemia-reperfusion injury(CIRI) and to explore the possible mechanisms on prevention and treatment for cerebral ischemia-reperfusion injury with Fufang Changpu Yizhi Decoction. Methods: Animal model of cerebral ischemiareperfusion injury was established by using the method of blocking bilateral common carotid artery. Radioimmunoassay was used to test the expression level of IL-1β in serum of mice of all groups. Immunohistochemisty was used to test the expression level of TGF-β1 in brain tissue of mice. Results: Compared with the sham operation group, the level of IL-1β and TGF-β1 in the model group increased signii cantly(P〈0.05). Compared with model group, the level of IL-1β of treatment groups decreased signii cantly(P〈0.05), but the level of TGF-β1 of each dose group increased signii cantly in brain tissue(P〈0.05), especially the high dose group of Fufang Changpu Yizhi Decoction(P〈0.05). Conclusion: Fufang Changpu Yizhi Decoction could reduce the degree of CIRI via adjusting the release of inflammatory factors to reduce inflammatory reaction so as to lower the intensity of cerebral ischemia-reperfusion injury.
