摘 要:目的:探讨中药复方固本防哮饮对哮喘易感基因血清类黏蛋白1样蛋白3(ORMDL3)蛋白表达的干预作用。方法:体内实验中,以卵蛋白(OVA)致敏小鼠为动物模型;体外实验中,以白介素-4(IL-4)和白介素-13(IL-13)刺激人胚肺成纤维细胞(MRC-5)为细胞模型,分别以固本防哮饮及其含药血清进行干预,通过肺组织病理评分评价动物造模用药前后的差异,Western Blot检测肺组织中及MRC-5细胞中ORMDL3蛋白含量的改变。结果:体内实验中,固本防哮饮能明显减轻肺组织气道炎性浸润(P<0.05),且显著下调ORMDL3蛋白的表达(P<0.05);体外实验中,固本防哮饮含药血清可下调MRC-5中ORMDL3蛋白表达(P<0.05)。结论:固本防哮饮改善哮喘患儿体质状态的药理机制之一可能在于调节易感基因ORMDL3蛋白的表达,从而有效减轻炎症反应,达到预防哮喘发作的目的。Objective: To discuss the effect of Guben Fangxiao Decoction on the expression of gene ORMDL3 susceptible to asthma. Methods: The experimental models such as ovalbumin(OVA) sensitized mice and human embryonic lungfibroblast(MRC-5) stimulated with interleukin-4(IL-4) and interleukin-13(IL-13) were all treated with Guben Fangxiao Decoction. Pathologic grading of lung tissue was used to evaluate the dif erence before and after establishing animal models and treating. Western Blot was used to test the expression of ORMDL3 in lung tissues and MRC-5 cells. Results: in vivo experiments, Guben Fangxiao Decoction could reduce the inflammatory infiltrates and ORMDL3 expression in lung tissue and airway obviously(P〈0.05). In vitro experiments, Guben Fangxiao Decoction could decrease the ORMDL3 protein expression in MRC-5 cells(P〈0.05). Conclusion: One of the mechanisms of Guben Fangxiao Decoction is the adjustment role of the expression of susceptibility gene ORMDL3. It can relieve inl ammation effectively to achieve the purpose of prevention of asthma.
