摘 要:目的:研究芎芍胶囊对动脉粥样硬化(AS)兔主动脉核转录因子κB(NF-κB)及相关炎症因子的影响,探讨其抗炎性反应抗AS的新靶点。方法:60只新西兰白兔随机分为空白对照组、模型组、芎芍小剂量、中剂量及大剂量组、辛伐他汀组,采用单纯高脂喂养法复制兔AS模型。给药前及给药6、12、15周后分别采集血样检测血清中白介素-2(IL-2)、白介素-6(IL-6)及肿瘤坏死因子-α(TNF-α)水平;15周后处死兔取胸主动脉采用非放射性凝胶迁移实验(EMSA)法测定其NF-κB p65活性表达,采用实时定量PCR测定NF-κB mRNA表达量。结果:造模给药15周后各组兔血清炎症因子均较前不同程度升高,芎芍中、大剂量组及辛伐他汀组较模型组IL-2、TNF-α明显减少(P<0.01,P<0.05);芎芍大剂量组、辛伐他汀组较模型组NF-κB p65结合活性降低(P<0.05)。结论:芎芍胶囊能够降低AS兔主动脉NF-κB p65的蛋白结合活性,减少血清中相关炎症因子,从而通过抑制炎性反应达到抗AS的目的。Objective: To study the effect of Xiongshao Capsule(XSC) on nuclear transcription factor κB(NF-κB) and inl ammation related factors of rabits with athrosclerosis(AS) and to investigate the new target for inhibiting the inflammatory response. Methods: Sixty New Zealand rabbits were randomly divided into control group, model group, low-dose XSC group, middle-dose XSC group, large-dose XSC group and simvastatin group. The rabbits of AS were established by simple high-fat feeding method. The levels of interleukin-2(IL-2), interleukin-6(IL-6) and the tumor necrosis factors-α(TNF-α) in the serum were tested by blood tests after remodeling, before delivery of drugs, after 6, 12, 15 weeks of delivery, separately. The activity expression of NF-κB p65 in thoracic aortas was tested by electrophoretic mobility shift assay(EMSA), and NF-κB mRNA was tested by Real-time quantitative PCR. Results: The serum inflammatory factors in medicated groups were increased to dif erent levels after 15 weeks. The IL-2 in middle-dose XSC group, high-dose XSC and simvastatin groups were lower than that in model group(P〈0.01, P〈0.05), and TNF-α were signii cantly lower(P〈0.05). The binding activity of NF-κB in simvastatin group was lower than that in model group(P〈0.05). Conclusion: The XSC can lower the binding activity of NF-κB p65 in aortas of rabbits with AS and reduce the serum inl ammation related factors, which could prevent AS through anti-inflammatory response.
