摘  要:目的:通过分析各组大鼠肺组织代谢物图谱,发现小青龙汤证肺组织的生物标志物,为临床诊断和治疗提供客观依据。方法:对空白对照组、模型组和小青龙汤组大鼠的肺组织进行核磁共振氢谱(1HNMR)检测,图谱经MestRec预处理和积分后,所得数据用SIMCA-P+进行PLS-DA分析,得到相应的散点图和载荷图,进行比较后找出差异性代谢物。结果:模型组与空白对照组相比,亮氨酸、异亮氨酸、缬氨酸、精氨酸、组氨酸、丙氨酸和牛磺酸增高(P<0.05),赖氨酸、苏氨酸和甲硫氨酸降低(P<0.05),醋酸无统计学意义。小青龙汤组与空白对照组、模型组相比,较空白对照组缬氨酸、精氨酸、牛磺酸、丙氨酸和组氨酸仍增高,较模型组缬氨酸、牛磺酸和丙氨酸有所降低,精氨酸比模型组中的含量还高;苏氨酸和甲硫氨酸较空白对照组低,较模型组高。结论:所找出的物质主要与能量代谢、炎症反应和免疫抵抗力有关,但与炎症及免疫相关的物质变化较明显。Objective: Through the map analysis of various metabolites in rats lung, to find out the biomarkers in syndrome suitable for Xiaoqinglong Decoction, and provide objective basis for the clinical diagnosis and treatment. Methods: Based on the rats lung NMR spectrum detection of the blank control group, model group and Xiaoqinglong Decoction group, the data from the map by MestRec pretreatment and integration were analyzed PLS-DA by using SIMCA-P+, then the corresponding scatter plot and loading diagram were got, and the dif erences of metabolites were found out by the comparison. Results: The results from comparison of blank control group and model group showed that the contents of metabolites of leucine, isoleucine, valine, arginine, histidine, alanine and taurine increased(P〈0.05), which of lysine, threonine and methionine decreased(P〈0.05), the acetic acid had no statistical signii cance. Compared with the blank control group and model group, Xiaoqinglong Decoction group showed that the contents of metabolites of valine, arginine, taurine, alanine and histidine were still higher than those in the blank control group, the contents of valine, taurine and alanine were lower than those in the model group, the contents of arginine was higher than those in the model group, the contents of threonine and methionine were lower than those in the blank control group but higher than model group. Conclusion: Through analyzing, these substances are mainly related to the energy metabolism, inflammatory response and immune resistance, but these substances which are relatied to inflammatory and immune changed obviously.
