雷氏消风祛湿巴布剂对大鼠佐剂性关节炎血清中IL-1β、PGE2水平的影响

摘　　要：目的探讨雷氏消风祛湿巴布剂对大鼠佐剂性关节炎血清中IL-1β、PGE2水平的影响。方法使用弗氏完全佐剂制作大鼠类风湿性关节炎模型,将大鼠分为空白对照组、模型组、阳性药物组(祛风骨痛巴布膏)、雷氏消风祛湿巴布剂低、中、高剂量组,比较各组大鼠足趾和关节肿胀度,并采用ELISA方法监测各组大鼠血清IL-1β、PGE2水平。结果造模第7天,各组大鼠足趾和关节肿胀度显著增加(P<0.05),经用药后,阳性对照组、雷氏消风祛湿巴布剂低、中、高剂量组大鼠足趾和关节肿胀度均有所缓解,其中各剂量巴布剂组随着剂量的增加,大鼠足趾和关节缓解程度越大;与空白组相比较,模型组、阳性对照组、雷氏消风祛湿巴布剂低、中、高剂量组大鼠血清IL-1β与PGE2水平均显著高于空白组(P<0.05);与模型组比较,各用药组(阳性对照组,低、中、高剂量组)大鼠血清IL-1β与PGE2水平均显著低于模型组(P<0.05),且各剂量组巴布剂大鼠,随着药物剂量的增加,血清IL-1β与PGE2水平显著降低。结论雷氏消风祛湿巴布剂能够显著改善大鼠佐剂性关节炎足趾和关节肿胀,其作用机制可能与抑制血清IL-1β、PGE2有关。OBJECTIVE To investigate the effects of Leishi Xiaofengqushi babu plaster on IL-1βand PGE2 Level in serum of adjuvant arthritis rats.METHODS Using Freund＇s complete adjuvant making rat rheumatoid arthritis model,the rats were divided into blank control group,model group,positive drug group（Qufenggutong babu plaster）,low,middle and high dose of Leishi Xiaofengqushi babu plaster groups to compare the swelling degree of rats＇ toes and joints.ELISA method was used to monitor the IL-1βand PGE2 level in the serum of rats in each group.RESULTS The rats toes and joints swelling degree increased significantly after 7days＇ molding（P〈0.05）.After the medication,rats＇ toes and joints swelling degree relieved in positive control group,and low,middle and high dose of Leishi Xiaofengqushi babu plaster groups.The greater the dose of all the babu plaster groups was,the greater the swelling degree of rats＇ toes and joints swelling was eased.Compared with the control group,the serum IL-1βand PGE2 level in model group,positive group,and Leishi Xiaofengqushi low,middle and high dose groups were significantly increased（P〈0.05）.Compared with the model group,the serum IL-1βand PGE2 level in positive group and Leishi Xiaofengqushi low,middle,and high dose groups were significantly lower than that in model group（P〈0.05）.The serum IL-1βand PGE2 level were significantly reduced with the increase of drug doses.CONCLUSION Leishi Xiaofengqushi babu plaster can significantly improve the swelling degree of rats＇ toes and joints.The mechanism might be related to the inhibition of serum IL-1β,and PGE2.
