摘  要:目的 探讨基于显微-CT(Micro-CT)及三维打印技术制备的仿生人类指骨支架复合脂肪干细胞(ADSCs)及重组人类骨形态蛋白2(rhBMP-2)后在体内的血管化。方法 取大白兔第3代ADSCs复合rhBMP-2接种到仿生指骨支架上混合培养10 d(实验组),和单纯仿生支架(对照组)分别包裹于12条成年比格犬腰背部两侧带蒂深筋膜瓣中,采用自身左右对照,不同时间进行大体、组织学染色观察比较新生血管。结果 两组支架材料在筋膜内均有一定的血管化能力,术后2、4、8、16周苏木素-伊红(HE)染色和免疫组织化学染色均显示实验组相对血管面积百分比和阳性染色的面密度值均优于同一时间的对照组,两者差异有统计学意义(P〈0.05)。结论 仿生人类指骨支架移植体内具备了血管化能力,复合ADSCs-rhBMP-2可以促进血管化程度。Objective Explore based on microscopic-CT and three-dimensional priming produce bionic human phalanx scaffold composite adipose-derived stem cells (ADSCs) and recombinant human bone morphogeuetic protein 2 (rhBMP-2) of blood vessels in the body, for the clinical treatment of phalangeal defects provide a theoretical basis Methods Take the 3rd generation ADSCs of the rabbits composite rhBMP-2, inoculated with Bionic phalanx bracket mixed culture after 10 days (experimental group) and simply Bionic phalanx bracket (control group) to wrapped in 12 adult beagle dogs lower back on both sides of the pedicle deep fascia, Use self-control, different times gross and histologic staining neovascularization Results The results shows two of the scaffolds in intrafascial both vascular capabilities. The amount of vessels and the expression of VIRRAg in experimental groups were stronger than those in control groups at 2, 4, 8, and 16 weeks, the difference was statistically significant (P 〈 0.05). Conclusion The bionic human phalanx scaffold transplant in body with vascular capacity, composite ADSCs-rhBMP-2 can contribute to the degree of vascularization.
