摘  要:目的研究地黄饮子对快速老化小鼠P8(SAMP8)学习记忆能力及海马组织病理形态的影响,探讨其治疗老年性痴呆的药效及作用机制。方法运用小鼠跳台行为学实验检测地黄饮子对SAMP8小鼠的学习记忆能力的影响;HE染色和电镜观察检测SAMP8小鼠海马形态结构和超微结构。结果模型组SAMP8小鼠跳台实验潜伏期缩短(第1天208.19±65.42,第2天254.76±48.99),与空白组SAMR1小鼠(第1天248.17±27.59,第2天296.35±3.17)比较有非常和/或显著差异(P<0.05,P<0.01)。给药后,地黄饮子组潜伏期(第1天277.06±7.86,第2天295.36±5.17)与模型组(第1天208.19±65.42,第2天254.76±48.99)比较非常显著性延长(P<0.01)。相同组的第2天潜伏期与第1天比较,除模型组以外,其他各组均非常显著延长(P<0.01)。模型组与空白组比较,伴有海马锥体细胞形态结构、超微结构的改变。给药后,地黄饮子减轻SAMP8小鼠的海马病理损伤状况。结论地黄饮子对SAMP8小鼠的学习记忆能力有改善作用。Objective To investigate the pharmacodynamic mechanisms of Di Huang Yin Zi( DHYZ) on learning and memory ability,neuropathology of senescence accelerated mouse prone 8. Methods The Step down test was used to detect the learning and memory ability of every mouse. HE staining and electron microscope were used for observing hippocampus neurons of structure and ultra structure. Results The escape latency of SAMP8 mice( 208. 19 ±65. 42,254. 76 ±48. 99) in step down test were shorter than the mice of SAMR1( 248. 17 ± 27. 59,296. 35 ± 3. 17),which were statistically significant( P0.05 or P0.01).After administration,compared with model group(208.19 ±65.42,254.76 ±48.99),DHYZ(277.06 ±7. 86,295. 36 ±5. 17) could increase the escape latency of SAMP8 mice in step down test,which were statistically significant( P〈0. 01). The results of the same groups except the model group were all extended between the different days( P〈0. 01). DHYZ could reduce the hippocampal pathological damage of SAMP8 mice. Conclusion It has a considerable improvement on the learning and memory ability of SAMP8 with DHYZ.
