Impact of Middle Components of Rhodiola Polysaccharide on Biochemical &Sperm Characteristics of Cryopreserved Boar Semen

摘  要:试验将采自5头杜洛克(2~4岁)猪精液(2次/周)冷冻解冻于含有红景天粗多糖中基分(Middle Components of Rhocliola Polysacchoride,MCRP)的稀释液中,应用计算机辅助精子分析系统对猪精子运动特征进行了评价,其它参数用常规精子评价方法进行评估,分析MCRP对猪精子冻后生化指标及活力的影响。结果表明,MCRP在稀释液中的有效浓度范围为4.0~6.0mg/L,当MCRP浓度为6mg/L时,其各项指标均显著高于其它各组(P〈0.05),而且冷冻解冻后,MCRP添加于稀释液I的精子质量显著优于MCRP添加到稀释液II的精子质量(P〈0.05)。冷冻解冻液中MCRP的浓度与解冻后解冻液中的谷胱甘肽、精子线粒体活性、精子低渗肿胀系数呈显著正相关(P〈0.05),而与解冻液中的丙二醛浓度呈显著负相关(r=-0.982,P〈0.05)。猪精液冷冻保护液中添加MCRP能够有效地抵抗由于冷冻产生的活性氧自由基对精子的伤害,从而提高精子冻后质量,但是MCRP抑制活性氧自由基的具体成分仍需要进一步的研究。Although being preliminary used as an antioxidant in cryopreserved boar semen, composition complexity of Rhodiola Polysaccharide deserves further study. In the study, middle Components of Rhodi- ola polysaccharide (MCRP) refined from Rhodiola Polysaccharide was used in cryopreserved boar semen and their effects on semen characteristics and its antioxidant properties were evaluated. Semen was collect- ed from five Duroc boars (2 to 4-yr-old,) twice weekly and frozen-thawed in extender with MCRP. Motion characteristics were assessed with a computer-aided semen analysis (CASA) system, and other sperm qual- ity end points were assessed by routine methods. The effective concentration of MCRP in extender ranged from 4 to 6 mg/L and all end points were significantly higher at 6 mg/L than in the control group(P〈0.05). More over the effect of MCRP on sperm quality was better in extender I than extender Ⅱ (P〈0.05). In frozen-thawed boar semen, there was a direct correlation (P〈0.05) between MCRP concentra- tion and glutathione concentrations (GSH), mitochondrial activity, and hypoosmotic swelling test (HOST), and an inverse correlation (r=0. 982,P(0.05) between MCRP concentration and malondial- dehyde. In summary, MCRP added in extender could effectively resist aggression that oxidative stress in cryopreservation harmed boar sperm. Further studies are needed to define the active ingredient in MCRP that protects boar sperm against ROS.
